摘要
目的:过敏性鼻炎(AR)动物模型是开展AR实验研究的基础,如何建立稳定可靠的AR动物模型是实验研究的关键。现有的AR动物模型种类繁杂多样,特点各异,结合文献挖掘对现有模型进行系统化分析,对其应用规律及潜在问题进行归纳整理,为建立稳定可靠的AR动物模型提供参考依据。方法:通过查阅相关资料,筛选出近十年AR动物模型相关文献,通过Excel表格建立数据库对其物种选择、模型类型、造模药物、基础致敏、激发方式、阳性药、检测指标等进行归纳整理和分析,整合各类模型的作用机理、模型特点,以及存在的问题。结果:在筛选出的366例动物模型中使用较多的动物物种为SD大鼠、Wistar大鼠、BALB/c小鼠和豚鼠;常用的基础致敏药物选用卵清蛋白(OVA),采用腹腔注射的方法,氢氧化铝作为佐剂(307例,83.88%);选用滴鼻激发(336例,91.81%)的激发方式;指标检测多从行为学评分、血清生化指标、鼻粘膜病理等多个方面开展。结论:在AR动物模型造模时建议使用SD大鼠、Wistar大鼠和BALB/c小鼠作为实验动物,采用滴鼻激发的方式,用OVA致敏,氢氧化铝作为佐剂,可以进一步提高模型的成功率,为建立稳定可靠的AR动物模型提供参考依据。
Objective: Allergic rhinitis(AR) animal model is the basis for conducting AR experimental research. To establish a stable and reliable AR animal model is the key to experimental research. The existing AR animal models are complicated, which have diversity and different characteristics. Combined with literature mining, the existing models were systematically analyzed, and their application rules and potential problems were summarized to provide a reference for establishing a stable and reliable AR animal model. Methods: By referring to relevant data, relevant documents of AR animal models in the past ten years were screened, and a database was established through EXCEL to summarize their species selection,the model type, modeling drugs, basic sensitization, excitation methods, positive drugs, test indicators, etc. Then the action mechanism, characteristic and existing problems of various models were organized and analyzed. Results: Among 366 animal models,SD rats, Wistar rats, BALB/c mice and guinea pigs were frequently used animal species. The commonly used basic sensitizing drug was ovalbumin(OVA), which were abdominally injected,and aluminum hydroxide was used as an adjuvant(307 cases, 83.88%), with nasal drip as the excitation method(336 cases, 91.81%). The frequently detected indexes were behavioral scores, serum biochemical indicators, nasal mucosal pathology, etc. Conclusion: It is recommended to use SD rats,Wistar rats and BALB/c mice as experimental animals in the AR animal model, nasal drop excitation, OVA sensitization,and aluminum hydroxide as an adjuvant. They can further improve the success rate of the models and provide reference for the establishment of stable and reliable AR animal models.
