过敏性哮喘患者外周血单个核细胞miR-138表达及与Th1/Th2平衡的关系 被引量:2

摘要 目的探讨过敏性哮喘患者外周血单个核细胞微小RNA-138(miR-138)表达及其与T辅助细胞1(Th1)/T辅助细胞2(Th2)平衡的关系。方法106例过敏性哮喘患者作为过敏性哮喘组(AAS),另取本院同期健康体检者100名作为对照组,荧光定量PCR法检测2组受试者外周血单个核细胞miR-138、Th1相关转录因子T-bet、Th2相关转录因子GATA3表达水平,酶联免疫吸附实验(ELISA)检测两组受试者Th1相关细胞因子白细胞介素-2(IL-2)、干扰素γ(IFN-γ)以及Th2相关细胞因子白细胞介素4(IL-4)、白细胞介素10(IL-10)含量。结果AAS组患者外周血单个核细胞miR-138表达水平显著高于对照组(P<0.05)。AAS组患者血清IL-2、IFN-γ含量显著低于对照组(P<0.05),IL-4和IL-10显著高于对照组(P<0.05);AAS组患者Th1/Th2比值显著低于对照组(P<0.05)。AAS组患者外周血单个核细胞中转录因子T-bet mRNA表达水平显著低于对照组(P<0.05),AAS组患者转录因子GATA3 mRNA显著高于对照组(P<0.05)。miR-138表达水平与细胞因子IL-2、IFN-γ呈负相关(r=-0.566、-0.616,P<0.05),与细胞因子IL-4、IL-10分别呈正相关(r=0.518、0.506,P<0.05)。miR-138表达水平与Th1相关转录因子T-bet mRNA水平呈负相关(r=-0.497,P<0.05),与Th2相关转录因子GATA3 mRNA水平呈正相关(r=0.653,P<0.05)。miR-138表达水平与Th1/Th2比值呈负相关(r=-0.606,P<0.05)。结论过敏性哮喘患者外周血单个核细胞miR-138表达水平上调,可能通过调节Th1/Th2影响相关细胞因子表达,影响过敏性哮喘患者机体炎症状态。 Objective To investigate the expression of micro RNA138(miR138)in peripheral blood mononuclear cells of patients with allergic asthma and its relationship with the balance of T helper cell 1(Th1)/T helper cell 2(Th2).Methods A total of 106 patients with allergic asthma were enrolled as allergic asthma group(AAS group,and 100 healthy subjects were enrolled as control group.Fluorescence quantitative PCR(PCR)was used to detect the expression levels of miR138,Th1-related transcription factor T-bet and Th2-related transcription factor GATA3 in peripheral blood mononuclear cells of the subjects in both groups.Moreover the levels of Th1-related cytokines interleukin2(IL-2),interferon-γ(IFN-γ)as well as Th2-related cytokines interleukin-4(IL-4)and interleukin-10(IL-10)were detected by enzyme-linked immunosorbent assay(ELISA).Results The expression levels of miR138 in peripheral blood mononuclear cells in AAS group were significantly higher than those in control group(P<0.05).The levels of IL-2 and IFN-γin AAS group were significantly lower than those in control group(P<0.05),however,the levels of IL-4 and IL-10 were significantly higher than those in control group(P<0.05).Moreover,the Th1/Th2 value and the expression levels of transcription factor Tbet mRNA in AAS group were significantly lower than those in control group(P<0.05),but levels of transcription factor GATA3 mRNA in AAS group were significantly higher than those in control group(P<0.05).Furthermore,the expression levels of miR138 were negatively correlated with those of cytokines IL-2 and IFN-γ(P<0.05),however,which were positively correlated with those of cytokines I-4 and IL-10(P<0.05).The expression levels of miR138 were negatively correlated with those of Th1-related transcription factor Tbet mRNA(P<0.05),however,which were positively correlated with those of Th2-related transcription factor GATA3 mRNA(P<0.05).In addition,the expression levels of miR138 were negatively correlated with Th1/Th2 value(P<0.05).Conclusion The expression levels of miR-138 in peripheral blood mononuclear cells of patients with allergic asthma are up-regulated,which may affect the expressions of related cytokines and the inflammatory state of the body in patients with allergic asthma by regulating Th1/Th2.
