摘要
目的研究金匮肾气丸对ICR老年痴呆小鼠模型保护作用研究。方法雄性ICR大鼠60只,体重32.5~35.5g,按体重随机分为5组,即:正常对照组、模型对照组、多奈哌齐组(3.0 mg/kg)及金匮肾气丸中、高(80mg/mL、120mg/mL)剂量组。正常对照组小鼠进行颈部皮下注射生理盐水(0.1ml/10g),其余组小水均进行造模(注射相同剂量的D-半乳糖),连续造模24天后进行新物体识别检测。将造模成功后的小鼠用随机数表法按照辨别指数重新分组,每组10只,连续给药24天后,进行新物体识别检测与Morris水迷宫检测,分析金匮肾气丸对老年痴呆小鼠的认知障碍样行为的保护作用。结果金匮肾气丸中、高剂量组小鼠能明显缩短潜伏期时间(P<0.05);金匮肾气丸中、高剂量组能明显提高老年痴呆小鼠的新物体辨别指数(P<0.05)结论金匮肾气丸能明显改善ICR老年痴呆模型的认知障碍样行为。
Research on the protective effect of Jinkui Shenqi Pills on ICR dementia mouse models.Methods:60 male ICR rats,weighing 32.5-35.5g,were randomly divided into 5 groups according to their body weights,namely:normal control group,model control group,Donepezil group(3.0 mg/kg)and Jinkui Shenqi Pill medium and high(80mg/mL,120mg/mL)dose group.The mice in the normal control group were injected with normal saline(0.1ml/10g)subcutaneously in the neck,and the other groups were modeled with small water(injected the same dose of D-galactose),and the new object was detected after 24 days of continuous model building.After the successful modeling,the mice were regrouped according to the discrimination index by the random number table method.Each group was 10 mice.After 24 days of continuous administration,the new object recognition test and Morris water maze test were performed to analyze the effect of Jinkui Shenqi Pill on Alzheimer’s disease.Protective effects of cognitive impairment-like behaviors in mice.Results The middle and high doses of Jinkui Shenqi Pills can significantly shorten the incubation period(P<0.05);the middle and high doses of Jinkui Shenqi Pills can significantly improve the new object discrimination index of Alzheimer’s mice(P<0.05)Conclusion:Jinkui Shenqi Pill can significantly improve the cognitive impairment-like behavior of the ICR Alzheimer’s model.
