重庆地区无偿献血者ABO亚型分布及鉴定浅析 被引量:7

Analysis on the distribution and identification of ABO subtypes of voluntary blood donors in Chongqing, China

摘要 目的对重庆地区无偿献血者ABO亚型分布情况和鉴定策略进行浅析。方法应用全自动血型分析仪对本中心2016年1月—2019年8月631 127份献血者标本进行ABO定型,对91例ABO血型正反不符合标本采用试管法、吸收放散等血清学方法检测,并对21例ABO正反定型不符合标本采用聚合酶链反应-序列特异性引物(PCR-SSP)分子生物学方法检测。结果血清学A亚型(A2,A3,Ael等),占比22.8%,B亚(Bx,Bw等)占比39.1%,A亚B(A2B等)占比9.8%, AB亚(cisAB, ABw,AB3等)占比28.3%。21例标本血清学方法与PCR-SSP方法检测结果符合率为47.7%。结论 ABO亚型需多种方法互相验证;ABO血型的准确鉴定,为安全输血提供有力的质量保障。 Objective To study the distribution of ABO subtypes in blood donors in Chongqing, so as to explore corresponding strategy of ABO subtype identification. Methods 91 samples with discrepant ABO blood typing results were screened out of 631 127 blood samples in our center from January 2016 to August 2019 using automatic blood typing analyzer. Such 91 samples suspected as weak blood group A and B phenotypes were serologically typed using glass tube method and absorption-elution tests. Genotyping for ABO using PCR-SSP was performed in 21 samples randomly selected from these 91 samples. Results Cases with phenotypic A subtypes(including A2, A3, Ael) was serologically found in 22.8%, B subtype(including Bx, Bw) in 39.1%, AsubB(including A2B) in 9.8%, and ABsub(including cisAB, ABw,AB3) in 28.3%. The blood types in 21 samples using serological test and PCR-SSP method were consistent in 47.7%. Conclusion Genotyping method would be a useful tool to assist serological test in discrepant ABO blood typing, which is helpful to improve accuracy in blood typing and safeguard the blood safety.
