大豆高蛋白基因分子标记辅助育种的应用 被引量:4

摘要 为了在周口地区大豆遗传背景的基础上筛选出有效的高蛋白基因分子标记,选用高蛋白大豆周豆25号和高油周豆18号作为亲本杂交构建F2分离群体,选择144对SSR引物,运用BSA方法进行大豆高蛋白基因SSR分子标记筛选,并利用Mapmaker/Exp V3.0软件构建分子连锁图谱和定位分析。结果表明,在亲本间具有多态性引物32对;这32对引物在高、低蛋白基因池间有4对引物(satt182、satt419、satt239、satt598)表现多态性,但这4对引物与蛋白相关的标记不连锁,其中,引物satt182贡献率最高(3.1%)。利用标记satt182对来自河南、北京等地的50份高蛋白大豆材料进行检测,验证分子标记辅助育种方法的有效性,结果显示,50份大豆材料中检测出38份与相应的特异性条带相同,检测符合度为76.0%,检测结果有效性较高、准确性较好。 To screen out effective molecular markers of high protein genes on the basis of soybean genetic background in Zhoukou area,high-protein soybean Zhoudou 25 and high-oil soybean Zhoudou 18 were selected as parental hybrids to construct F2 segregation population.144 pairs of SSR primers were selected,SSR molecular markers of soybean high-protein genes were screened by BSA method,and Mapmaker/Exp V3.0 software was used to construct the molecular linkage map and location analysis.The results showed that there were 32 pairs of primers that were polymorphic between the parents.These 32 pairs of primers showed polymorphisms in 4 pairs of primers(satt182,satt419,satt239 and satt598)between the high and bottom protein pools(BSA method).But these 4 pairs of primers were not linked to the protein-related markers.Among them,the contribution rate of primer satt182 was the highest,which was 3.1%.The marker satt182 was used to detect 50 samples of high-protein varieties from Henan,Beijing and other places to verify the effectiveness of the molecular marker-assisted breeding method.The result showed that thirty-eight copies were detected that were identical to the corresponding specific bands,and the detection agreement was 76.0%.The detection results were effective and accurate.
