小麦品种西农979抗赤霉病基因的转录组测序鉴定 被引量:1

Identification of FHB Resistance Genes in Wheat Cultivar Xinong 979 by Transcriptome Sequencing

摘要 小麦品种西农979对赤霉病具有较好的抗性。为了发掘西农979的抗赤霉病基因,本研究采用禾谷镰刀菌菌株BN-8分别接种抗病品种西农979与感病品种周麦18,取接种前西农979颖壳及接种后12、24和96 h的西农979与周麦18颖壳进行转录组分析,筛选抗感赤霉病差异表达基因,并对其进行GO功能注释及KEGG富集分析。结果表明,接种后12、24和96 h各出现14、1978和142个差异表达基因,共计2122个基因。这些基因经GO功能注释分类分成3大类43个亚类,主要涉及代谢过程、细胞过程、应激反应、生物调控、细胞部件、细胞、催化活性、结合、转运活性等。KEGG通路分析结果显示,内质网中的蛋白质加工通路中差异基因最多,有48个;其次是光合作用—天线蛋白通路及淀粉和蔗糖代谢通路,各含41和30个差异基因;显著富集的通路有光合作用—天线蛋白、内质网中的蛋白质加工、萜骨架的生物合成等。植物激素信号转导通路中发现27个差异基因,参与脱落酸和茉莉酸等路径;植物-病原体互作通路中发现19个差异基因,编码钙结合蛋白、WRKY转录因子、抗病蛋白等。此外,还筛选到UDP-葡萄糖基转移酶基因等脱毒相关蛋白基因。 The wheat cv.Xinong 979 has good resistance to FHB.In order to explore FHB disease resistance genes in Xinong 979,Xinong 979 and a susceptible wheat cv.Zhoumai 18 were inoculated by Fusarium graminearum strain BN-8.The glumes of the two cultivars at 12,24 and 96 h after inoculation and the glumes of Xinong 979 before inoculation were collected for transcriptome sequencing to screen differentially expressed genes(DEGs)related to FHB resistance.GO function annotation and KEGG pathway analysis were carried out for the DEGs.The results showed that there were 2122 DEGs,including 14,1978 and 142 DEGs at 12,24 and 96 h after inoculation,respectively.GO function annotation classified these DEGs into 43 functional groups of three major classes,mainly involving metabolic process,cellular process,response to stimulus,biological regulation,cell part,cell,catalytic activity,binding and transporter activity.KEGG pathway analysis showed that protein processing in endoplasmic reticulum enriched the most DEGs(48),followed by photosynthesis-antenna proteins,containing 41 DEGs and 30 DEGs annotated in starch and sucrose metabolism,respectively.The pathways significantly enriched were photosynthesis-antenna proteins,protein processing in endoplasmic reticulum,and terpenoid backbone biosynthesis,etc.There were 27 DEGs participated in plant hormone signal transduction,involved in abscisic acid pathway,jasmonic acid pathway and so on.19 DEGs were found in plant-pathogen interaction,encoding calcium-binding proteins,WRKY transcription factors,disease resistance proteins,and so on.In addition,we also screened detoxification related protein genes,such as UDP-glucosyltransferase genes.
