广藿香长链非编码RNA分析

Analysis on Long Non-coding RNAs in Pogostemon cablin(Blanco)Benth.

摘要 长链非编码RNA(lncRNA)是一类长度大于200个核苷酸的非编码RNA,能够在表观遗传水平、转录水平和转录后水平调控基因表达,广泛参与植物的生长发育、抗逆、生理和病理过程。广藿香为广东道地药材之一,也是医药领域和化工领域的重要天然原料。为了更好地开发和利用这一重要资源,本研究以广藿香为实验材料,在全长转录组测序的基础上,利用四个软件对134647个转录本全长序列进行了蛋白编码潜能预测,最终获得4106个lncRNAs。对这些lncRNAs的长度和数量作统计分析。结果显示,最短为280 nt,最长为8941 nt,平均长度为1084 nt;大部分lncRNAs的长度介于280~1000 nt(61.447%),在1001~2000 nt和2001~4000 nt的长度范围内也发现有较多的lncRNAs,分别为23.015%和15.270%;但在4001~8941 nt的长度范围内,总共只有11个lncRNAs(占总量的0.268%)。本研究为广藿香lncRNAs鉴定和后续的功能分析提供了重要的基础。 Long chain non-coding RNA(lncRNA)is a non-coding RNA of more than 200 nucleotides in length.LncRNAs play important roles in such a wide range of biological proceses such as growth,development,physiology,pathology and streses by regulating gene expresion at epigenetic,transcription and post-transcription level.Pogostemonis Herba(Pogostemon cablin(Blanco)Benth.)is one of the authentic medicinal materials in Guangdong Province and an important natural raw material in the field of medicine and chemical industry.In order to better explore and utilize the important resource,our study based on the full-length transcriptome sequencing of P.cablin to predict the protein coding potential of 134647 transcripts by four softwares,and finally obtained 4106 lncRNAs.Statistical analysis of the length and number of these lncRNAs showed that the shortest was 280 nt,the longest was8941 nt,and the average length was 1084 nt.Most lncRNAs length ranged from 280 to 1000 nt(61.447%).Within the length range of 1001~2000 nt and 2001~4000 nt,more lncRNAs were found,which were 23.015%and15.270%,respectively.But in the range of 4001~8941 nt,there were only 11 lncRNAs in total(0.268%).The results provided an important basis for the identification and subsequent functional analysis of P.cablin lncRNAs.
