病毒性心肌炎患者血浆IL-17A表达及IL-17A基因多态性分析 被引量:5

Expression of plasma IL-17A in patients with viral myocarditis and IL-17A gene polymorphism

摘要 目的探究病毒性心肌炎(Viral myocarditis,VMC)患者白细胞介素-17A(Interleukin-17A,IL-17A)表达及IL-17A基因多态性与病情易感性的关系。方法选择2017年1月-2019年3月佛山市中医院收治的VMC患者60例,根据患者病程长短分为VMC急性期组36例和VMC恢复期组24例,选择同期于佛山市中医院体检的30名健康者作为对照组。采用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术检测IL-17A基因rs2275913(G-197A)位点多态性;采用酶联免疫吸附(ELISA)法检测血清IL-17A水平。结果三组研究对象rs2275913(G-197A)位点多态性分布符合遗传平衡检验(P<0.05);VMC急性期组和VMC恢复期组AA、GG基因型频率与对照组比较,差异有统计学意义(P<0.05);VMC急性期组和VMC恢复期组的A、G等位基因频率与对照组比较,差异有统计学意义(P<0.05);以AG基因型为参照,经logistic回归分析矫正混杂因素(性别、年龄等),IL-17A基因rs2275913(G-197A)位点携带AA基因型及A等位基因患者VMC发病风险分别为AG基因型的2.108倍和2.056倍。VMC急性期组和VMC恢复期组患者血清IL-17A水平显著高于对照组(P<0.05);AA、AG基因型血清IL-17A水平显著高于GG基因型(P<0.05)。结论血清IL-17A在病毒性心肌炎的发生、发展中发挥重要作用;IL-17A基因rs2275913(G-197A)位点多态性可能与病毒性心肌炎易感性有关,A等位基因可能为易感基因。 OBJECTIVE To explore the expression of interleukin-17 A(IL-17 A)in patients with viral myocarditis(VMC)and observe the correlation between IL-17 A gene polymorphism and susceptibility to the disease.METHODS A total of 60 patients with VMC who were treated in Foshan Hospital of Traditional Chinese Medicine from Jan 2017 to Mar 2019 were enrolled in the study and divided into the acute phase of VMC group with 36 cases and the VMC recovery group according to the course of the disease,meanwhile,30 healthy people who received physical examination in Foshan Hospital of Traditional Chinese Medicine were set as the control group.The polymorphism of IL-17 A gene rs2275913(G-197 A)was detected by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP),and the serum IL-17 A level was detected by means of enzyme-linked immunosorbent assay(ELISA).RESULTS The distribution of polymorphism of rs2275913(G-197 A)loci of the three groups of study objects conformed to the genetic balance test(P<0.05).There were significant differences in the frequencies of AA and GG genotypes between the acute phase of VMC group,the VMC recovery group and the control group(P<0.05).There were significant differences in the frequencies of A and G alleles between the acute phase of VMC group and the VMC recovery group(P<0.05).Taken the AG genotype as a reference,and confounding factors such as gender and age were corrected by logistic regression analysis,the patients who had the IL-17 A gene rs2275913(G-197 A)locus carrying with AA genotype and the patients who had the IL-17 A gene rs2275913(G-197 A)locus carrying with A allele were respectively 2.108 times and 2.056 times the risk of VMC as the patients who had AG genotype.The serum IL-17 A of the acute phase of VMC group and the VMC recovery group was significantly higher than that of the control group(P<0.05).The serum IL-17 A level of the patients who had AA and AG genotypes were significantly higher than that of the patients who had GG genotype(P<0.05).CONCLUSION The serum IL-17 A plays an important role in the occurrence and development of VMC.The polymorphism of IL-17 A gene rs2275913(G-197 A)loci may be associated with the susceptibility to VMC,and A allele may be a susceptible gene.
