Simultaneous Determination of Four Flavonoids in Huaihua Powder by HPLC

摘要 目的建立HPLC同时测定槐花散中芦丁、柚皮苷、新橙皮苷、槲皮素4种黄酮类成分含量的方法。方法采用依利特Hypersil ODS2色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL/min,检测波长283 nm,柱温25℃。结果芦丁、柚皮苷、新橙皮苷、槲皮素线性范围分别为0.2526~25.26μg(R2=1.0000)、0.1555~15.55μg(R2=1.0000)、0.1005~10.05μg(R2=1.0000)、0.0208~2.08μg(R2=0.9998),平均加样回收率分别为101.2%(RSD=1.4%)、99.4%(RSD=2.2%)、102.5%(RSD=1.9%)、98.7%(RSD=2.7%)。结论本研究建立的含量测定方法简便、准确,重复性好,可用于槐花散的质量控制。 Objective To establish an HPLC method for the simultaneous determination of rutin,naringin,neohesperidin and quercetin in Huaihua Powder.Methods The analysis was performed on a 25℃thermostatic Elite Hypersil ODS2 column(250 mm×4.6 mm,5μm),with the mobile phase comprising of acetonitrile-0.1%phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 283 nm.Results The linear range of four components was:rutin,0.2526‒25.26μg(R2=1.0000);naringin,0.1555‒15.55μg(R2=1.0000);neohesperidin,0.1005‒10.05μg(R2=1.0000);quercetin,0.0208‒2.08μg(R2=0.9998).The average recoveries of four components were:rutin,101.2%(RSD=1.4%);naringin,99.4%(RSD=2.2%);neohesperidin,102.5%(RSD=1.9%);quercetin,98.7%(RSD=2.7%).Conclusion The method is simple and accurate,with good repeatability,which can be used for the quality control of Huaihua Powder.
