南蛇藤提取物靶向mTOR抑制人胃癌细胞SGC-7901的迁移能力的研究 被引量:5

摘要 目的研究不同浓度的南蛇藤Celastrus orbiculatus Thunb.提取物对敲低雷帕霉素靶蛋白(Mammalian Target of Rapamycin,mTOR)表达的人胃癌SGC-7901细胞迁移能力的影响。方法用siRNA技术,构建敲低mTOR表达的人胃癌SGC-7901细胞。MTT法检测南蛇藤提取物(10、20、40、80、160、320μg·mL^-1)对人胃癌SGC-7901/mTOR-细胞增殖的影响。加入不同浓度的南蛇藤提取物(20、40、80μg·mL^-1)作用24 h后,分别用划痕实验和Transwell小室迁移实验,考察不同浓度的南蛇藤提取物对SGC-7901/mTOR-细胞迁移能力的影响。Western blot法分析对mTOR信号转导通路相关蛋白的影响。结果南蛇藤提取物明显抑制人胃癌SGC-7901/mTOR-细胞的增殖,呈浓度依赖性。南蛇藤提取物明显降低人胃癌SGC-7901/mTOR-细胞的迁移能力,减少mTOR信号转导通路相关蛋白的表达量。结论南蛇藤提取物明显抑制人胃癌SGC-7901/mTOR-细胞的迁移,其分子机制可能与mTOR信号转导通路相关。 Objective To study the effects of different concentrations of Celastrus orbiculatus Thunb.Extracts on the migration ability of human gastric cancer SGC-7901 cells knocking down the expression of Mammalian Target of Rapamycin(mTOR).Methods Human gastric cancer SGC-7901 cells with knockdown of mTOR expression were constructed using siRNA technology.The MTT method was used to detect the effects of the extract of C.orbiculatus(10,20,40,80,160,320μg·mL^-1)on the proliferation of human gastric cancer SGC-7901/mTOR-cells.After adding different concentrations of extracts of C.orbiculatus(20,40,80μg·mL^-1)for 24 h,the scratch test and Transwell cell migration experiment were used to investigate the effect of different concentrations of C.orbiculatus extract on SGC-7901/mTOR cell migration ability.Western blot was used to analyze the effects of mTOR signal transduction pathwayrelated proteins.Results The extract of C.orbiculatus significantly inhibited the proliferation of human gastric cancer SGC-7901/mTOR-cells in a concentration-dependent manner.The extract from C.orbiculatus significantly reduced the migration capacity of human gastric cancer SGC-7901/mTOR-cells,and reduced the expression of proteins related to the mTOR signal transduction pathway.Conclusion The extract of C.orbiculatus significantly inhibits the migration of human gastric cancer SGC-7901/mTOR-cells,and its molecular mechanism may be related to the mTOR signal transduction pathway。
