摘要
采用试验方法,对白及种子不同贮藏方法以及白及苗培育进行了研究,结果表明在相同的贮藏时间,含水率为5%、温度为4℃时白及种子的活力和萌发率最高;在3/4MS培养基中添加0.7 mg/L NAA、0.2 mg/L 6-BA,白及种子萌发率最高,种胚变成绿色的时间最短。在3/4 MS培养基中,添加0.5 mg/L 6-BA+0.3 mg/L NAA,假鳞茎诱导率最高,其直径、根数、根长均达到了差异显著水平。基于以上结果,生产上白及种子贮藏最适宜的条件为含水率5%,贮藏温度4℃;白及种子萌发的最佳培养基为3/4 MS+0.7 mg/L NAA+0.2 mg/L 6-BA+土豆粒70 g/L+蔗糖30 g/L+琼脂6.5 g/L+活性炭0.3 g/L;适宜白及假鳞茎诱导、植株生长发育的最佳培养基为3/4 MS+0.5 mg/L 6-BA+0.3 mg/L NAA+香蕉50 g/L+土豆70 g/L+蔗糖30 g/L+琼脂7 g/L+活性炭0.3 g/L。
The effects of different treatments on seed germination rate,and pseudobulb induction rate of Bletilla to provide theoretical guidance and experimental basis for seed storage and pseudobulb induction.The results showed that the seed vigor and germination rate were the highest,when the moisture content was 5%and the temperature was 4℃.The seed germination rate was the highest and the colour of seeds changed from brown to green in the shortest time,when 0.7 mg/L NAA and 0.2 mg/L 6-BA were added in 3/4 MS medium.On the basis of 3/4 MS Medium,0.5 mg/L 6-BA+0.3 mg/LNAA was added,and the induction rate of pseudobulb was the highest,and the diameter of pseudobulbs,the number and the length of roots were significantly different from other treatments.In conclusion,the most suitable conditions for the storage of Bletilla seeds were moisture content of 5%and a storage temperature of 4%.The optimal medium for the seed germination was 3/4 MS+0.7 mg/L NAA+0.2 mg/L 6-BA+mashed potatoes 70 g/L+sucrose 30 g/L+agar 6.5 g/L+activated Carbon 0.3 g/L.The best medium for pseudobulbs induction and plant growth and development of Bletilla was medium 3/4 MS+0.5 mg/L 6-BA+0.3 mg/L NAA+banana 50 g/L+potato70 g/L+sucrose 30 g/L+agar 7 g/L+activated carbon 0.3 g/L.
