摘要 目的在体外条件下研究宫炎平片(GYP)是否具有遗传毒性,为临床用药安全提供依据。方法对GYP设不同剂量组,选用Ames试验、体外CHL细胞染色体畸变试验和小鼠骨髓嗜多染红细胞微核试验进行遗传毒性试验。结果Ames试验各组平皿均未见细菌毒性表现,结果为阴性。CHL试验中24 h后采样以及48 h后采样,所有剂量的细胞染色体畸变率均未显著增高,GYP无抑制有丝分裂或诱导染色体数目畸变的能力,微核试验结果为阴性无统计学意义(P>0.05)。结论综合以上3个试验结果,认为在实验条件下,GYP无遗传毒性。 OBJECTIVE To investigate the genetic toxicity of Gongyanping Tablet(GYP),in order to provide useful data for clinical drug safety.METHODS Ames test,CHL chromosome aberation assay and micronucleus assay of bone marrow cell in mice were used to test the genotoxicity of different doses of GYP.RESULTS No bacterial toxicity was found in the dishes of each group in Ames test,and the results were negative.No significant increase of chromosome aberration rate was observed in 24 h and 48 h after CHL test.No mitotic inhibition or induction of chromosome number aberration was observed in GYP.The results of micronucleus test showed that there was no significant difference between the two groups(P>0.05).CONCLUSION Based on the results of the above three tests,it is concluded that GYP has no genotoxicity under the conditions of our laboratory.
