夹江石斛水提取物对金黄色葡萄球菌的抑菌作用及机制研究 被引量:5

Studies on the Antibacterial Activity and Mechanism of the Water Extracts from Dendrobium jiajiangense on Staphylococcus aureus

摘要 探讨夹江石斛水提取物(DJWEs)对金黄色葡萄球菌的抑菌作用及抑菌机制。采用96孔板微量肉汤稀释法以及平板培养法检测DJWEs对金黄色葡萄球菌的最小抑菌浓度(MIC),并通过测定培养液中碱性磷酸酶(AKP)、电导率、大分子物质吸光度(OD)值以及菌体DAPI的荧光强度,研究DJWEs对金黄色葡萄球菌细胞壁通透性、细胞膜通透性和核酸合成的影响。DJWEs对金黄色葡萄球菌的MIC浓度为70 mg/mL;经DJWEs处理后,金黄色葡萄球菌培养液中AKP的含量明显增高(P<0.05),但电导率和大分子物质的OD值差异无统计学意义(P>0.05),而DAPI染色后核酸荧光强度有显著变化(P<0.01)。DJWEs对金黄色葡萄球菌有较强的抑菌作用,抑菌机理可能涉及影响金黄色葡萄球菌的细胞壁通透性和核酸DNA和RNA合成。 To study the antibacterial activity and mechanism of the Dendrobium jiajiangense water extracts(DJWEs)on Staphylococcus aureus.The 96-well plate 2-fold dilution method was adopted to determine the minimal inhibitory concentrations(MIC).The detection of alkaline phosphatase(AKP),electrical conductivity and OD value,and DAPI staining assay were used to primarily study the antibacterial mechanism of DJWEs against Staphylococcus aureus.The MIC was 70 mg/mL.After treated with DJWEs,the AKP and DAPI staining of Staphylococcus aureus had significant differences.The significant antibacterial activity of DJWEs was found on Staphylococcus aureus.The antibacterial mechanism of DJWEs may involve in influencing cell wall permeability and nucleic acid including DNA and RNA synthesis of Staphylococcus aureus.
