补肾疏肝汤对慢性疲劳综合征大鼠模型抗疲劳作用机制的实验研究 被引量:4

摘要 目的研究补肾疏肝汤对慢性疲劳综合征大鼠模型抗疲劳的作用机制。方法将50只SD大鼠按照随机数字表法分为5组,即空白组、模型组及补肾疏肝汤低、中、高剂量组,各组均10只。除空白组外,其余4组大鼠均制备慢性疲劳综合征模型。在造模同时,空白组、模型组均予0.9%氯化钠注射液40 m L/(kg·d)灌胃;补肾疏肝汤低、中、高剂量组分别予补肾疏肝汤40 m L/(kg·d)灌胃,生药含量分别为17.28、34.56、69.12 g/(kg·d),分别相当于人体用量1、2、4倍,均14 d。比较各组大鼠第1 d首次、第14 d末次力竭游泳时间。比较各组大鼠血清超氧化物歧化酶(SOD)、丙二醛(MDA)水平,下丘脑5-羟色胺(5-HT)含量,大鼠骨骼肌过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)、细胞色素氧化酶(COX)含量。结果补肾疏肝汤低、中、高剂量组大鼠第14 d末次力竭游泳时间均较模型组延长(P<0.05),且补肾疏肝汤中剂量组大鼠第14 d末次力竭游泳时间最长(P<0.05)。补肾疏肝汤低剂量组第14 d末次力竭游泳时间较第1 d首次力竭游泳时间缩短(P<0.05);补肾疏肝汤中剂量组第14 d末次力竭游泳时间较第1 d首次力竭游泳时间延长(P<0.05)。模型组大鼠血清SOD水平低于空白组(P<0.05),MDA水平高于空白组(P<0.05)。补肾疏肝汤低、中、高剂量组大鼠血清SOD水平均高于模型组(P<0.05),补肾疏肝汤低、中剂量组血清MDA水平低于模型组(P<0.05)。补肾疏肝汤中剂量组大鼠血清SOD水平高于补肾疏肝汤低、高剂量组(P<0.05),MDA水平低于补肾疏肝汤低、高剂量组(P<0.05)。模型组大鼠下丘脑5-HT含量低于空白组(P<0.05)。补肾疏肝汤中、高剂量组大鼠下丘脑5-HT含量均高于模型组(P<0.05),且补肾疏肝汤中剂量组高于补肾疏肝汤高剂量组(P<0.05)。模型组大鼠骨骼肌PGC-1α、COX含量均低于空白组(P<0.05)。补肾疏肝汤低、中、高剂量组大鼠骨骼肌PGC-1α、COX含量均高于模型组(P<0.05),且补肾疏肝汤中剂量组大鼠骨骼肌PGC-1α、COX水平最高(P<0.05)。结论补肾疏肝汤能增强慢性疲劳综合征大鼠模型抗疲劳作用,补肾疏肝汤中剂量组更明显。 Objective To explore the anti fatigue mechanism of Bushen Shugan decoction on chronic fatigue rats.Methods 50 SD rats were divided into five groups according to the random number table method,namely blank group,model group and low,middle and high dose group of Bushen Shugan decoction,with 10 rats in each group.In addition to the blank group,the other four groups of rats were prepared for chronic fatigue syndrome model.After the successful establishment of the model,the blank group and the model group were given 40 mL/(kg·d)sodium chloride injection by gavage.In the low,middle and high dose groups of Bushen Shugan decoction,they were given 40 mL/(kg·d)Bushen Shugan decoction respectively,and its crude drug content was 17.28,34.56 and 69.12 g/(kg·d)respectively,which was 1,2 and 4 times of the human dosage.The time of the first and last exhaustive swimming was recorded.The levels of serum superoxide dismutase(SOD)and malondialdehyde(MDA)in each group were compared.The contents of 5-HT in hypothalamus in each group were observed.The levels of peroxisome proliferator activated receptorγcoactivator 1α(PGC-1α)and cytochrome oxidase(COX)were observed.Results Compared with the model group,the last exhaustive swimming time of rats in the low,middle and high dose group of Bushen Shugan decoction was longer(P<0.05),and the middle dose group of Bushen Shugan decoction was the longest(P<0.05).The last exhaustive swimming time of rats in the 14 d in the low dose group of Bushen Shugan decoction was shorter than that in the first day of the first exhaustive swimming time(P<0.05).The last exhaustive swimming time of rats in the 14 d in the middle dose group of Bushen Shugan decoction was shorter than that in the first day of the first exhaustive swimming time(P<0.05).The serum SOD of the model group was lower than that of the blank group(P<0.05),and the MDA of the model group was higher than that of the blank group(P<0.05).The serum SOD of the low,middle and high dose group of Bushen Shugan decoction was higher than that of the model group(P<0.05),and the MDA of the low and middle dose group of Bushen Shugan decoction was lower than that of the model group(P<0.05).The serum SOD in the middle dose group was higher than that in the low and high dose group of Bushen Shugan decoction(P<0.05),and the MDA was lower than that in the low and high dose group of Bushen Shugan decoction(P<0.05).The level of 5-HT in the hypothalamus of the model group was lower than that of the blank group(P<0.05).The level of 5-HT in hypothalamus of rats in the middle and high dose groups of Bushen Shugan decoction was higher than that in the model group(P<0.05),and the middle dose group was higher than the high dose group of Bushen Shugan decoction(P<0.05).The levels of PGC-1αand Cox in the skeletal muscle of the model group were lower than those of the blank group(P<0.05).The levels of PGC-1αand Cox in the low,middle and high dose groups of Bushen Shugan decoction were higher than those in the model group(P<0.05),and the levels of PGC-1αand COX in the skeletal muscle of middle dose group of Bushen Shugan decoction were the highest(P<0.05).Conclusion Bushen Shugan decoction can enhance the anti fatigue effect of chronic fatigue syndrome rat model.The middle dose group is more worthy of clinical application.
