摘要
生物样本库近年由于在生物医学领域中的重要作用获得快速发展。本文综述了低温保存技术在生物样本库建设中的应用现状。在低温环境中保存不可避免会对生物样本产生损伤,分析了低温保存损伤生物样本的机制,包括添加保护剂会损伤细胞、降温复温过程中的机械损伤以及低温保存对生物大分子的影响。并结合现阶段生物样本库对低温保存技术的要求提出了改进方法的研究方向,对未来生物样本库应用低温保存技术的发展趋势进行了展望。
The biobank has been developed rapidly in recent years due to its important role in the field of biomedicine.The application status of cryopreservation technology in the construction of biobanks is reviewed in this article.Damage to biological samples is unavoidable when stored in a low temperature environment,then the mechanism of damaging biological samples by cryopreservation is analyzed,including cell damage caused by the addition of cryoprotectant,mechanical damage during cooling and rewarming,and the effects of cryopreservation on biological macromolecules.Based on the current requirements of biobanks to cryopreservation technology,the research direction of improving methods is put forward,and the development trend of the application of cryopreservation technology in biobanks is prospected.
