摘要
目的探讨聚多巴胺纳米颗粒(DOP)表面修饰腺样囊性癌细胞表面表达的叶酸(FA)受体的配体后,对腺样囊性癌的靶向作用及在近红外光下发挥光热治疗作用。方法多巴胺经氧化形成聚多巴胺纳米颗粒,再连接巯基化的叶酸得到表面修饰叶酸的DOP(DOPFA)。随后对纳米材料的尺寸、电位以及形貌进行评价,再经红外分析颗粒成分,考察该体系的升温性能以及肿瘤细胞杀伤能力以及肿瘤细胞靶向能力。结果 DOPFA具有较小的尺寸(~240nm),具有良好的升温性能,能高效地进行光热杀伤肿瘤细胞。体外肿瘤细胞吞噬实验表明,该体系有较好的腺样囊性癌细胞靶向能力。结论 DOPFA在腺样囊性癌的靶向治疗方面具有很好的应用前景。
Objective To investigate the targeting and photothermal effect of DOP on adenoid cystic carcinoma by modifying the ligand of folic acid(FA)receptor expressed on the surface of adenoid cystic carcinoma cells.Methods Dopamine was first oxidized to form polydopamine nanoparticles and then FA was conjugated onto nanoparticles(DOPFA)through Michael reaction.Size and Zeta potential were tested and morphology was observed via TEM,following by determining temperature property,tumor cell killing ability,and tumor cell targeting ability.Results The DOPFA has an average size of 240 nm,which has good temperature rising performance,and can efficiently kill tumor cells by photothermal treatment.Besides,in vitro tumor cell phagocytosis experiments show that DOPFA has good targeting ability for adenoid cystic carcinoma cells.Conclusion DOPFA has a good prospect in targeting therapy of adenoid cystic carcinoma.
