摘要
目的:探究益肺方含药血清对肺腺癌A549细胞增殖的影响。方法选取15只大耳兔分别给予高剂量益肺方、低剂量益肺方和生理盐水连续灌胃14天处理,采心脏血液并取其血清,并在体外培养的肺癌A549细胞株中加入上述益肺方含药血清。使用MTT法检测肺腺癌A549细胞增殖能力,使用Annexin V/PI法检测细胞凋亡、使用Westen Bolt和RT-PCR定性、半定量检测Bcl-2、Bax、P-mTOR、P-AMPK、caspase-3、caspase-9的表达量。结果MTT比色结果显示,相比空白对照组,低剂量组和高剂量组细胞抑制率显著升高(P<0.01),相比低剂量组,高剂量组的抑制率显著提高(P<0.01);相比空白组,低剂量组和高剂量组含药血清caspase-3、caspase-9表达量显著增加,相比低剂量组,高剂量组caspase-3、caspase-9表达量显著增加(P<0.01);相比空白组,低剂量组和高剂量组Bcl-2、P-mTOR表达量显著降低,Bax、P-AMPK表达量显著增加且两者有显著性差异(P<0.01),相比低剂量组,高剂量组Bcl-2、P-mTOR表达量显著降低,Bax、P-AMPK表达量显著增加且两者有显著性差异(P<0.01)。结论:益肺方含药血清可以抑制肺腺癌A549细胞增殖,同时可以通过调控AMPK/mTRO信号通路促进结直肠癌细胞的凋亡。
Objective:To explore the effect of serum-containing Yifei Recipe on the cell proliferation of lung adenocarcinoma A549.Methods:15 rabbits were selected and treated with high-dose Yifei Recipe,low-dose Yifei Recipe and normal saline for 14 days of continuous gavage.The blood of heart was taken and serum was taken,and the above-mentioned serum-containing Yifei Recipe was added to the lung cancer A549 cell line cultured in vitro.MTT assay was used to detect the proliferation of lung adenocarcinoma A549 cell.Apoptosis was detected by Annexin V/PI method.The expression levels of Bcl-2,Bax,P-mTOR,P-AMPK,caspase-3 and caspase-9 were detected qualitatively and semi-quantitatively by Westen Bolt and RT-PCR.Results:MTT colorimetric results showed that compared with the blank controlled group,the cell inhibition rates in the low-dose group and the high-dose group were significantly increased(P < 0.01).Compared with the low-dose group,the inhibition rate was significantly increased in the high-dose group(P < 0.01).Compared with the blank group,the expression levels of serum-containing caspase-3 and caspase-9 were significantly increased in the low-dose group and the high-dose group,and the expression levels of caspase-3 and caspase-9 in the high-dose group were significantly increased compared with those in the low-dose group(P < 0.01).Compared with the blank group,the expression levels of Bcl-2 and P-mTOR were significantly decreased in the low-dose group and the high-dose group,and the expression levels of Bax and P-AMPK were significantly increased(P < 0.01).Compared with the low-dose group,the expression levels of Bcl-2 and P-mTOR were significantly decreased in the high-dose group,and the expression levels of Bax and P-AMPK were significantly increased(P < 0.01).Conclusion:Serum-containing Yifei Recipe can inhibit the proliferation of lung adenocarcinoma A549 cell,and can promote the apoptosis of colorectal cancer cells by regulating AMPK/mTRO signaling pathway.
