摘要
目的建立高效液相色谱法(HPLC)同时测定莨菪浸膏片中东莨菪碱、山莨菪碱、硫酸阿托品和东莨菪内酯的含量。方法采用Welch C1(8150 mm×4.6 mm,5μm)色谱柱,甲醇-0.05%磷酸溶液梯度洗脱,流速1.0 mL/min,进样量20μL,检测波长215 nm。结果4种组分分别在0.501~20.059μg/mL,0.988~19.761μg/mL,0.468~9.361μg/mL和0.502~10.030μg/mL浓度范围内线性关系良好,平均回收率在98.3%~100.0%之间,RSD小于2.8%。结论该方法简单准确,灵敏度高,可用于莨菪浸膏片真伪辨析和质量控制。
Objective To establish a high performance liquid chromatography(HPLC)method for simultaneous determination of scopolamine,anisodamine,atropine sulphate and atropine sulphate in belladonna extract tablets.Methods HPLC separations were carried out on a Welch C18 column(150 mm×4.6 mm,5μm)with a linear gradient elution using methanol and 0.05%phosphoric acid at a flow rate of 1.0 mL/min.The injection volume was 20μL and the detection wavelength was 215 nm.Results Good linear relationships between the peak area and concentration were found in the ranges of 0.501~20.059μg/mL,0.988~19.761μg/mL,0.468~9.361μg/mL and 0.502~10.030μg/mL for four components,respectively.The average recoveries were in the ranges of 98.3%~100.0%with the RSD less than 2.8%.Conclusion The method is simple,accurate and sensitive.It can be used for distin guishing the false and quality control of the belladonna extract tablets.
