摘要
目的研究不同炮制方法对不同产地续断中的马钱苷酸、川续断皂苷乙及川续断皂苷Ⅵ的含量变化和抗氧化活性的影响。方法采用HPLC法分别测定四川、云南、贵州及湖北产地的续断经微波炮制后,生、炒、盐及酒炙品中马钱苷酸、川续断皂苷乙及川续断皂苷Ⅵ的含量。Agilent C 18色谱柱(250 mm×4.6 mm,5μL);乙腈-1 mL·L^-1磷酸溶液进行梯度洗脱;流速:1.0 mL·min^-1;检测波长:254 nm;柱温:30℃;进样量:10μL。同时采用紫外分光光度计以DPPH·清除率来评价其抗氧化活性。结果云南炒续断马钱苷酸含量最高,湖北酒炙川续断皂苷乙含量最高,四川酒炙川续断皂苷Ⅵ含量最高;湖北产地生续断、炒续断及盐炙续断抗氧化活性强于其他产地;四川产地酒炙续断抗氧化活性最强。结论不同产地不同炮制方法均能影响续断中马钱苷酸、川续断皂苷乙及川续断皂苷Ⅵ的含量及抗氧化活性。
Objective To study the effect of various processing methods on the content change of loganic acid,dipsacoside B and dipsacosideⅥin Radix Dipsaci from different areas and on the antioxidation activity.Methods The contents of loganic acid,dipsacoside B and dipsacosideⅥin raw,frying,salty and winny Radix Dipsaci after microwave processing from Sichuan,Yunnan,Guizhou and Hubei areas,were determined by HPLC with an Agilent C 18 column(250 mm×4.6 mm,5μL)and a mobile phase of acetonitrile-1 mL·L^-1 phosphoric acid solution for gradient elution.The flow rate was 1.0 mL·min^-1 and the detection wavelength was 254 nm.The column temperature was 30℃,and the injection volume was 10μL.The clearance rate of DPPH·was used to evaluate the antioxidant effect by UV spectrophotometer.Results The content of loganic acid in frying product was much higher than the others from Yunnan.The content of dipsacoside B in wine product was much higher than the others from Hubei.The content of dipsacosideⅥin wine product was much higher than the others from Sichuan.The antioxidant activity of the raw,frying and salty products from Hubei was much higher than the others.The antioxidant activity of the wine product from Sichuan was the highest.Conclusion Different areas and different processing methods have effect on the content and antioxidation of loganic acid,dipsacoside B and dipsacosideⅥin Radix Dipsaci.
