摘要
目的研究支原体菌落形成单位(cfu)和颜色改变单位(ccu)2种微生物定量方法计数支原体的定量关系。方法用A8培养基肉汤、支原体培养基肉汤及SP4培养基肉汤五倍梯度对应稀释ATCC33175、ATCC23114及ATCC29342冻干粉,再对应接种A8琼脂培养基、支原体琼脂培养基及SP4琼脂培养基,培养后按ccu和cfu方法对支原体计数。结果cfu计量3株支原体标准差都比ccu计量大,计量数值也都比ccu计量大,ccu和cfu计量ATCC33175差异无统计学意义(P>0.05),而二者计量ATCC23114和ATCC29342差异有统计学意义(P<0.05)。结论采用五倍梯度稀释的ccu计量,ATCC33175无差异,其他脲原体是否有同样结果仍需要进一步研究,通过降低梯度稀释倍数是否可以消除2种方法计量ATCC23114和ATCC29342产生的显著性差异也需要进一步研究。
Objective To study the quantitative relationship between two microbial quantitative methods such as colony formation unit(cfu)and color changing unit(ccu)for counting mycoplasma.Methods The five times-fold gradient of A8 broth,mycoplasma broth and SP4 broth correspondingly diluted ATCC33175,ATCC23114 and ATCC29342 freeze-dried powder,and then correspondingly inoculated A8 agar,Mycoplasma agar and SP4 agar.After culture,the mycoplasma was counted according to the ccu and cfu methods.Results The standard deviation measured by cfu was larger than that measured by ccu,and the measurement values were also larger than that measured by ccu.The difference between the measurement of ccu and cfu in ATCC33175 was not statistically significant(P>0.05),while the differences between the measurement of ccu and cfu in ATCC23114 and ATCC29342 were statistically significant(P<0.05).Conclusion The difference in ATCC33175 was not statistically significant when ccu was measured with five-fold gradient dilution.Whether other ureaplasma was the same needs further study,and whether the significant differences in ATCC23114 and ATCC29342 measured by two methods could be eliminated by reducing the gradient dilution ratio also needs further study.
