摘要
目的:通过对益气温阳方对变应性鼻炎小鼠NK细胞亚群NK1型细胞以及NK2型细胞细胞因子IL-5、IL-10、IL-4、INF-γ分化影响的研究,初步探讨益气温阳方对活化后NK细胞的作用机制。方法:将30只C57BL/6小鼠随机分为空白组、模型组和益气温阳方低、中、高剂量组及西替利嗪组,共6组,每组5只;以卵白蛋白致敏方法建立AR模型后,空白组、模型组灌胃生理盐水,益气温阳方低、中、高剂量组分别灌服益气温阳方低、中、高3种不同浓度的水煎液,西替利嗪组灌服西替利嗪水溶液;给药后分别制备6组含药血清;取正常小鼠,处死分离脾脏,分离纯化原代NK细胞,分别用准备好的6组含药和空白血清制备NK细胞混悬液,除空白组外,其余加入尘螨试剂,用Elisa法测定NK细胞内的IL-5、IL-10、IL-4、INF-γ的表达情况。结果:变应性鼻炎小鼠NK2型偏倚可能主要是由于IL-4的增多;益气温阳方低、中、高剂量均不同程度的抑制了活化后的NK2型细胞释放IL-4、IL-5,并不同程度的促进了活化后的NK1型细胞释放IL-10,其中益气温阳方中剂量组效果最佳;西替利嗪组抑制NK2型细胞释放IL-4、IL-5效果同益气温阳方中剂量相似,但促进活化后的NK1型细胞释放IL-10效果高于中药组;西替利嗪组、中药组对NK1型细胞INF-γ的释放无明显促进作用。结论:益气温阳方可以抑制NK2型细胞细胞因子的释放,促进NK1型细胞因子的表达,提示益气温阳方治疗变应性鼻炎的机制可能与其抑制NK2型细胞活化,促进NK1型细胞活化有关。
Objective:To explore the action mechanism of Yiqi Wenyang Formula on activated NK cells by studying the effects of Yiqi Wenyang Formula on differentiations of IL-5,IL-10,IL-4 and INF-γfrom NK1 and NK2 cells in mice with allergic rhinitis(AR).Methods:30 C57BL/6 mice were randomly divided into the blank group,the model group,Yiqi Wenyang Formula groups of low-dose,medium-dose and high-dose,as well as Cetirizine group,with 5 mice in each group.After AR model was established by sensitization with ovalbumin,each group was given corresponding intervention.Drug-containing serum was prepared after medication.Normal mice were sacrificed,spleen was separated and primary NK cells were purified.NK cell suspension was prepared with drug-containing serum and blank serum respectively.Except blank group,the rest were added dust mite reagent,and Elisa method was used to detect the expressions of IL-5,IL-10,IL-4 and INF-γin NK cells.Results:It showed that the increase of IL-4 may be the main cause of NK2 bias in mice with allergic rhinitis.Different concentrations of Yiqi Wenyang Formula inhibited the release of IL-4 and IL-5 from activated NK2 cells in varying degrees,and promoted the release of IL-10 from activated NK1 cells in varying degrees,of which the medium dose had the best effect.The effect of Cetirizine was similar to medium-dose of Yiqi Wenyang Formula on inhibiting the release of IL-4 and IL-5 from activated NK2 cells;however,it was superior to Yiqi Wenyang Formula groups in promoting IL-10 release from activated NK1 cells.Both Cetirizine and Yiqi Wenyang Formula had no obvious effect on the release of INF-γfrom NK1 cells.Conclusion:Yiqi Wenyang Formula can inhibit the cytokines release from NK2 cells and promote the cytokines expressions of NK1 cells,which shows that the action mechanism of Yiqi Wenyang Formula in treating allergic rhinitis may be related to inhibiting the activation of NK2 cells and promoting the activation of NK1 cells.
