摘要
目的建立HPLC测定人工关节假体周围感染患者关节液中万古霉素浓度的方法。方法以替硝唑为内标,采用HypersilC18柱(250mm×4.6mm,5μm),流动相为乙腈-0.0125mol·L^-1磷酸二氢钾缓冲液(pH=3.2)(10∶90),流速1 mL·min^-1,柱温35℃,检测波长236nm,进样量10μL。结果关节液中万古霉素浓度在0.9068~90.6818μg·mL^-1线性良好。批内、批间精密度RSD均<15%,准确度85.89%~103.51%。结论本法易于操作,精密度和准确度良好,适用于人工关节假体周围感染患者关节液中的万古霉素浓度监测。
OBJECTIVE To establish a method for determination of vancomycin concentration in synovial fluid of patients with prosthetic joint infection by HPLC.METHODS Used tinidazole as internal standard substance,vancomycin concentration in synovial fluid was analyzed by Hypersil C18(250 mm×4.6 mm,5μm)with mobile phase of acetonitrile-0.0125 mol·L^-1 potassium dihydrogen phosphate buffer solution(pH=3.2)(10∶90).The flow rate was 1 mL·min^-1.The column temperature was 35℃and the injection volume was 10μL.RESULTS Vancomycin concentration in synovial fluid was in well linearity in the range of 0.9068?90.6818μg·mL^-1.The relative standard deviations of intra-batch and inter-batch precision was<15%,and the accuracy was between 85.89%^-103.51%.CONCLUSION The method is simple,sensitive and accurate,which is suitable for monitoring of vancomycin concentration in synovial fluid of patients with prosthetic joint infection.
