摘要
以7株芽孢杆菌为研究对象,对其主要生物学特性进行了研究,包括抑制大肠杆菌能力、产酶特性、对人工胃液和人工肠液的耐受性。结果表明,巨大芽孢杆菌、侧孢芽孢杆菌具有较强的抑制大肠杆菌的能力,枯草芽孢杆菌的蛋白酶总活力最高,淀粉酶活力最高的是巨大芽孢杆菌,脂肪酶活力最高的是地衣芽孢杆菌,纤维素酶活性最高的是枯草芽孢杆菌,7株芽孢杆菌均对人工胃液的耐受性较强,除地衣芽孢杆菌、侧孢芽孢杆菌外,其他5株芽孢杆菌对人工肠液的耐受性较强。通过本次研究综合分析,巨大芽孢杆菌在7株菌中的抑制大肠杆菌能力最强,产酶能力及耐人工胃液、肠液的能力也较强,具有作为新型饲料微生态制剂的开发潜力。进一步采用紫外诱变的方法提高该株巨大芽孢杆菌的抑制大肠杆菌的能力。结果显示:最佳的诱变时间为80 s,诱变后抑菌率为90.92%,比出发菌株增加了6.16个百分点,5次传代试验结果表现出较好的遗传稳定性,适合用于新型微生态制剂产品的研发。
The main biological characteristics of 7 strains of Bacillus were studied,including the abili?ty to inhibitE.coli,the characteristics of enzyme production,and the tolerance to artificial gastric juice and intestinal juice.The results showed thatBacillus megaterium andBacillus lateralis had strong abil?ity to inhibit E.coli,Bacillus subtilis had the highest total protease activity,Bacillus megaterium had the highest amylase activity,Bacillus licheniformis had the highest lipase activity,Bacillus subtilis had the highest cellulase activity and 7 strains had the highest cellulase activity.Bacillus had strong toler?ance to artificial gastric juice.Except Bacillus licheniformis and Bacillus lateralis,the other five strains of Bacillus had strong tolerance to artificial gastric juice.Through the comprehensive analysis of this study,Bacillus megaterium has the strongest ability to inhibit E.coli among the seven strains,and has the stronger ability to produce enzymes and to tolerate artificial gastric juice and intestinal juice.It has the potential to develop as a new feed microecological preparation.Ultraviolet mutagene?sis was used to improve the ability ofBacillus megatus to inhibitE.coli.The results showed that the best mutagenesis time was 80 s,the rate of inhibition was 90.92%,which was 6.16 percentage points higher than the original strain.The results of 5 passages showed good genetic stability,which was suitable for the research and development of new microecological preparations.
