抗褐化剂对水曲柳成熟胚外植体褐化及体胚发生的影响 被引量:2

Effects of Anti-browning Agents on Explant Browning and Somatic Embryogenesis from Mature Zygotic Cotyledons of Fraxinus mandshurica Rupr.

摘要 利用抗坏血酸(ASA)、谷胱甘肽(GSH)、硝酸银(AgNO3)和聚乙烯吡咯烷酮(PVP)等抗褐化剂处理,检验水曲柳(Fraxinus mandshurica Rupr.)外植体褐化与体胚发生的响应格局及其相互关系。结果表明:①10 mg·L^-1的ASA处理显著降低了植体褐化指数(代表外植体褐化面积比值)、褐化率和体胚发生率;随着ASA质量浓度的增加,褐化指数、褐化率和体胚发生率不断增加;在100 mg·L^-1时,褐化率达到98%(等同于对照),褐化面积和体胚发生率分别达到0.734和73%,均显著高于对照(P<0.05);200 mg·L^-1对外植体致死;协方差与相关性分析表明,ASA处理对水曲柳外植体褐化与体胚发生的影响格局之间具有显著相关性。②GSH对水曲柳外植体褐化指数和褐化率的影响较小,无规律性变化格局;10、20 mg·L^-1的低质量浓度处理显著降低体胚发生率,但随质量浓度增加体胚发生率增加,在200 mg·L^-1处理时显著高于对照,与100 mg·L^-1以下ASA处理的体胚发生影响趋势一致。③AgNO3在显著降低水曲柳外植体褐化指数和褐化率的同时也显著降低了体胚发生率;PVP处理对水曲柳外植体褐化影响不大,各处理体胚发生率虽均高于对照,但无规律性变化格局。④GSH、PVP和AgNO3这3种抗褐化剂对水曲柳外植体褐化与体胚发生的影响格局之间不具有显著相关性。综上,ASA可以用来进行水曲柳体胚发生伴随外植体褐化现象生物学机制解析的优选试剂。 Ascorbic acid(ASA), glutathione(GSH), silver nitrate(AgNO3) and polyvinylpolypyrrolidone(PVP) were used as anti-browning agents to test the response patterns of explant browning, somatic embryogenesis and their relationship for Fraxinus mandshurica Rupr.. The results showed that:(1)The explant browning index(represents the rate of browning area on explants), browning percentage and somatic embryo(SE) induction rate were significantly decreased in the treatment of 10 mg·L^-1 ASA, they were gradually increased along with the increase of ASA treating concentrations and reached 98% of browning percentage(equals to control), 0.734 and 73% of browning index and SE induction rate respectively(significantly higher than those of control), but lethal to explants in 200 mg·L^-1 ASA treatment;covariance and correlation analysis indicated that there was correlation between explant browning and somatic embryogenesis of the species.(2)There was little effect of GSH on browning index and percentage, i.e., no regular change patterns of the two indexes;the SE induction rate were significantly reduced in 10 and 20 mg·L^-1 low concentration treatments, it was increased along with the GSH concentration increase and significantly higher than that of control in 200 mg·L^-1 treatment;the changing pattern was similar to that of ASA in treatments below 100 mg·L^-1 treatments.(3)The explant browning and SE induction of F. mandshurica were significantly reduced in AgNO3 treatments at the same time;there was no evident influence of PVP treatments on the explant browning of F. mandshurica and no regular influencing pattern but with higher rate for SE induction by PVP treatments.(4)By covariance and correlation analysis, there was no correlation between explant browning and somatic embryogenesis of the species treated by AgNO3, GSH and PVP. The results implied that ASA could be a preference agent for further elucidating the biological mechanism of the concomitant phenomenon of somatic embryogenesis and explant browning of F. mandshurica.
