摘要
目的观察丹皮酚(Paeonol)对大肠癌细胞增殖、凋亡及自噬的影响,并探讨其可能的作用机制。方法用不同浓度的丹皮酚(0、30、60、120mg/L)在体外处理大肠癌LoVo细胞,分别用CCK-8比色法、PI/Annexin V双染法、蛋白质印迹法检测LoVo细胞活力、细胞凋亡率、细胞凋亡相关蛋白(Bax和Bcl-2)及自噬相关基因蛋白(Atg5、LC3-Ⅱ和Beclin1)的表达情况。结果通过丹皮酚处理后,CCK-8比色法显示丹皮酚以剂量、时间依赖的方式抑制LoVo细胞的增殖;PI/Annexin V双染后流式细胞仪检测显示,随着药物浓度的增加,LoVo细胞凋亡率也逐渐增加;免疫印迹结果显示,LoVo细胞凋亡相关蛋白Bax蛋白表达增加,抗凋亡相关蛋白Bcl-2蛋白的表达降低;此外,丹皮酚能上调LoVo细胞自噬相关基因Atg5、LC3-Ⅱ和Beclin1蛋白的表达。结论丹皮酚对大肠癌LoVo细胞有明显的抗增殖和促凋亡作用,其机制可能是通过诱导细胞自噬实现的。
Objective To observe the effects of paeonol on the cell proliferation,apoptosis and autophagy in colorectal cancer cells and explore the possible mechanisms.Methods The LoVo cells were treated with paeonol at different concentrations(0,30,60 and 120mg/L)respectively at different time.The proliferaton of LoVo cells was measured by CCK-8 assay.Apoptosis was detected by PI/Annexin V stainning.Western blot were used for Bax,Bcl-2,Atg5,LC3-Ⅱand Beclin1 protein analysis.Results From the data of CCK-8,the cell proliferation of LoVo cells was inhibited by paeonol in a dose-dependent and time-dependent manner.After treated with paeonol,the apoptosis rate of LoVo cells increased.The treatment with paeonol promoted the expression of pro-apoptotic factor Bax,and suppressed the expression of anti-apoptotic factor Bcl-2.Paeonol could promoted the expressions of Atg5,LC3-Ⅱand Beclin1 protein.Conclusion Paeonol could inhibit the cell proliferation and induce the cell apoptosis of LoVo cells,which may be associated with promotion of autophagy.
