摘要
目的:通过小动物分子影像技术及实验检测结果,研究储存红细胞输注对细菌感染引发的肝损伤作用。方法:在实验小鼠尾静脉注射细菌200μl,感染每只实验小鼠,将400μl红细胞悬液(RBCs)通过异氟烷麻醉小鼠的眼后静脉丛输注。在细菌感染小鼠模型中采用小动物分子影像技术观察细菌体内成像的结果,检测新鲜和储存RBCs输注造成铜绿假单胞菌诱导感染小鼠的肝损伤及肝切片的组织学检查结果。结果:根据细菌体内成像的结果,显示注射的铜绿假单胞菌在小鼠的肝脏区域诱导最高强度的荧光素酶信号,而且储存RBCs输注在体内增强了铜绿假单胞菌诱导的肝损伤;与新鲜RBCs相比,用储存RBCs处理的小鼠中丙氨酸转氨酶(ALT)和谷草转氨酶(AST)血清水平显著升高,与肝切片的组织学检查显示一致的结果。结论:储存RBCs的输注增强铜绿假单胞菌诱导感染小鼠的体内肝损伤,加剧脓毒血症的严重程度。
Objective:To study the effect of the infusion of stored red blood cells on liver injury caused by bacterial infection through molecular imaging technology of small animal and experimental test results.Methods:Each experimental mouse was infected with 200μl of bacteria which were injected into their tail vein,and 400μl red blood cell suspension(RBCs)was infused through the posterior eye venous plexus of mice which were anesthetized by isoflurane.In the mouse model with bacterial infection,molecular imaging technique of small animal was used to observe the results of intracorporeal imaging of bacteria.And the results of histological examination of liver injury and liver sections of P.aeruginosa-infected mice that were caused by infusion of fresh and stored RBCs were detected.Results:Based on the results of in vivo imaging for bacteria,it was shown that the injected P.aeruginosa could induce luciferase signal with the highest intensity in the liver region of mice,and that the infusion of stored RBCs strengthened P.aeruginosa-induced liver injury in vivo.Compared with that of fresh RBCs,the serum levels of ALT and AST in mice that were treated with stored RBCs were significantly increased.This results showed consistence with that of histological examination of liver sections.Conclusion:The infusion of stored RBCs can enhance liver injury in mice infected with P.aeruginosa and can exacerbate the severity of sepsis.
