摘要
目的探讨长链非编码RNA H19和E2F转录因子1(E2F transcription factor 1,E2F1)在子宫内膜癌(EC)中的表达情况及临床意义。方法收集64例EC患者的EC组织及其相应的癌旁组织标本,采用实时荧光定量聚合酶链反应(qRT-PCR)检测EC组织及癌旁组织中H19、E2F1的表达情况,采用免疫组化SP法检测EC组织及癌旁组织中E2F1蛋白的表达情况,分析EC组织中H19、E2F1的表达情况与患者临床特征及预后的关系,比较EC组织中二者表达的相关性。结果H19和E2F1在EC组织中的相对表达量明显高于癌旁组织,且EC组织中E2F1蛋白的阳性表达率明显高于癌旁组织(P﹤0.01)。不同年龄、分化程度、国际妇产科联盟(FIGO)分期EC患者EC组织中H19的高表达情况比较,差异均无统计学意义(P﹥0.05);不同组织学分级、淋巴结转移情况EC患者EC组织中H19的高表达情况比较,差异均有统计学意义(P﹤0.05)。不同年龄EC患者EC组织中E2F1的高表达情况比较,差异无统计学意义(P﹥0.05);不同FIGO分期、分化程度、组织学分级、淋巴结转移情况EC患者EC组织中E2F1的高表达情况比较,差异均有统计学意义(P﹤0.05)。Pearson相关性分析结果显示,EC患者EC组织中H19和E2F1的表达呈正相关(r=0.750,P﹤0.01)。H19和E2F1低表达的EC患者的生存情况分别优于H19和E2F1高表达的EC患者(P﹤0.05)。结论H19和E2F1在EC患者的EC组织中均表达上调且呈正相关,并与患者的某些临床特征有关,可能协同参与EC的发生、发展,有潜力成为预测EC患者预后的潜在标志物。
Objective To investigate the expression and clinical significance of long non-coding RNA H19 and E2F transcription factor 1(E2F1)in endometrial carcinoma(EC).Method Sixty-four EC patients were included in the study,from whom the EC tissues and tissues surrounding tumor were collected,the expression levels of H19 and E2F1 in all samples were detected by quantitative real-time-polymerase chain reaction(qRT-PCR),besides,E2F1 protein expression in EC and tissues surrounding tumor were detected using immunohistochemical SP assay,the relationship of H19 and E2F1 with patients’clinical characteristics and prognosis were analyzed,and the correlation between them in EC tissues was further investigated.Result The relative expression of H19 and E2F1 in EC tissues was significantly higher than that in tissues surrounding tumor,additionally,the positive expression rate of E2F1 protein in EC tissues was significantly higher than that in tissues surrounding tumor(P<0.01).There was no significant difference regarding the high expression of H19 in EC tissues from EC patients in terms of age,degree of differentiation and International Federation of Gynecology and Obstetrics(FIGO)stage,with no significant differences noted(P>0.05),while there was statistically significant differences regarding the high expression of H19 in EC tissues from EC patients in terms of histological grade and lymph node metastasis(P<0.05).The high expression of E2F1 in EC tissues of EC patients of different ages was not statistically significant(P>0.05).Comparison of the high expression of E2F1 in EC tissues of EC patients with different FIGO stage,degree of differentiation,histological grade,and lymph node metastasis showed statistically significant differences(P<0.05).Pearson correlation analysis indicated that,there was a positive correlation between the expressions of H19 and E2F1 in EC patients(r=0.750,P<0.01).The survival of EC patients with low expression of H19 and E2F1 was better than that of those with high expression of H19 and E2F1(P<0.05).Conclusion H19 and E2F1 are all up-regulated in EC tissues of EC patients,and there exists a positive correlation between them,furthermore,both markers are related to certain clinical characteristics of patients,indicating their possible involvement in the occurrence and development of EC,therefore implicating their potential values of serving as predictors for the prognosis of EC patients.
