摘要
为研究全缘叶绿绒蒿的ISSR遗传多样性,本研究以全缘叶绿绒蒿16个居群共185个个体为研究对象,采用简单序列重复区间扩增多态性(ISSR)标记技术分析全缘叶绿绒蒿遗传多样性,利用Popgen1.32计算遗传相似系数(GS)、Shannon指数(I)、Nei’s指数(He)等遗传参数;AMOVA进行遗传变异分析;利用MEGA4.0软件及UPGMA法进行聚类分析并构建树状图,并用MVSP中的主成分分析(PCA)对群体的遗传关系进行聚类分析;用TFPGA软件进行Mental检测。ISSR的21条引物共扩增出279个条带,平均每条引物扩增出13.3个条带,其中多态性条带278个,条带在150~2 000 bp之间均有分布,物种水平上的遗传多样性百分比PPB=99.64%,Nei’s基因多样性指数(He)范围在0.052 0~0.208 3之间,Shannon’s多样性信息指数(I)在0.079 5~0.313 4范围内;遗传分化系数(Gst)和AMOVA分析均显示遗传变异主要存在于种群间。种群间基因流(Nm)小于1,遗传漂变在全缘叶绿绒蒿的遗传分化中所起作用较大。Mental相关性检验,p值均大于0.05,说明遗传距离与地理距离间相关性不显著,地理距离对居群间的遗传分化没有明显影响。通过全缘叶绿绒蒿的遗传多样性研究发现,ISSR是一种非常便捷、有效的分子标记技术,能够为后续的种质鉴定、杂交育种和资源收集与合理开发利用提供一定的理论依据和参考。
To study the genetic diversity of Meconopsis integrifolia by ISSR. A total of 185 individuals from 16 populations of M. integrifolia were studied. Analysis of genetic diversity of M. integrifolia by using simple sequence repeat region amplified polymorphic(ISSR) markers. Using Popgen1.32 to calculate genetic similarity coefficient(GS), Shannon Index(I), Nei’s Index(He) and other genetic parameters. Analysis of genetic variation by AMOVA. Using MEGA4.0 software and UPGMA method to cluster analysis and construct tree diagram.Cluster analysis of population genetic relationship using principal component analysis in MVSP(PCA). Mental detection by TFPGA Software. A total of 279 bands were amplified from 21 primers of ISSR, with an average of13.3 bands per primer, of which 278 were polymorphic bands, all of which were distributed between 150 and2 000 bp. Percentage of genetic diversity at species level is 99.64%(PPB=99.64%). The Nei’s gene diversity index(He) ranges from 0.052 0 to 0.208 3. The Shannon’s diversity information index(I) is in the range of 0.079 5~0.313 4. Genetic differentiation coefficient( Gst) and AMOVA analysis showed that genetic variation mainly existed between populations. Gene flow(Nm) between populations is less than 1, which indicates that genetic drift plays an important role in genetic differentiation of M. integrifolia. Mental correlation test showed that p values were all greater than 0.05, indicating that there was no significant correlation between genetic distance and geographical distance, and geographical distance had no significant effect on genetic differentiation among populations. Based on the genetic diversity of M. integrifolia, ISSR is a convenient and effective molecular marker technique. ISSR can provide some theoretical basis and reference for further germplasm identification, cross breeding, resource collection and rational development and utilization.
