摘要
优化根瘤菌(Rhizobium NG10)产多糖的培养条件,提高胞外多糖(extracelluar polysaccharides of Rhizobium,REPS)产量,并探究其抗肿瘤活性。通过单因素实验、Plackett-Burman实验和Box-Behnken实验等对根瘤菌产胞外多糖发酵培养基配方进行优化,并采用MTT法检测多糖对人肝癌细胞的抑制作用。最终优化得到的最优培养基配方为:麦芽糖22.5 g/L,大豆蛋白胨9.5 g/L,MgSO 4·7H 2O 0.2 g/L,KH 2PO 4 0.41 g/L,NaCl 0.1 g/L,此时胞外多糖产量为5.05 g/L,相比原始提高了45.95%;REPS对肝癌细胞Hep G2的抑制呈一定的量效关系,培养时间为48 h时IC 50为393.3μg/mL,肝癌细胞抑制率达到56.74%。
In this study,the optimal the culture conditions for production of extracellular polysaccharides by Rhizobium sp.NG10 were determined.Besides,the inhibitory effect of the polysaccharides on human hepatocellular carcinoma cells was studied by MTT assay.The optimal medium composition for extracellular polysaccharides production were 22.5 g/L maltose,9.5 g/L soybean peptone,0.2 g/L MgSO 4·7H 2O,0.41 g/L KH 2PO 4 and 0.1 g/L NaCl.Under these conditions,the yield of the polysaccharides was increased 45.95%with titer of 5.05 g/L.Moreover,the inhibitory effect of the polysaccharides on human hepatocellular carcinoma cells exhibited a dose-effect relationship.When the human hepatocellular carcinoma cells were cultured with the polysaccharides for 48 h,the IC 50 was 393.3μg/mL,and the inhibition rate was 56.74%.
