摘要
制备山羊乳酪蛋白单克隆抗体可以为检测乳品中山羊乳成分提供免疫学检测手段。以山羊酪蛋白免疫Balb/c小鼠,应用细胞融合技术,制备特异性识别山羊乳酪蛋白的单克隆抗体,对获得的阳性杂交瘤细胞进行连续传代和反复冻存以测定其稳定性,并对其分泌的单克隆抗体应用ELISA进行抗体特异性分析,对特异性识别山羊乳酪蛋白的单克隆抗体进行抗体亚型鉴定和染色体组型分析。最终,成功的筛选出可以分泌特异性识别山羊乳酪蛋白的单克隆抗体的阳性杂交瘤细胞株,被命名为7H。经染色体组型分析,染色体数目为102条;单克隆抗体7H类别为IGg1,轻链为κ;经连续传代和反复冻存,ELISA检测细胞上清抗体发现其OD 450nm值无显著性差异。
The preparation of monoclonal antibody against goat can provide immunological detection methods for the detection of goat milk in dairy products.Therefore,in this study,we immunized Balb/c mice with goat casein,and cell fusion technology was used to screen out monoclonal antibodies that specifically recognize goat casein.The obtained hybridoma cells were serially passaged and repeatedly cryopreserved.The secreted monoclonal antibodies were analyzed by ELISA for antibody specificity,and then antibody isoforms and karyotype analysis tests were performed.Finally,we successfully screened the monoclonal antibodies against goat casein,which was named 7H.
