不同基因型大豆耐盐性筛选与综合鉴定 被引量:12

摘要 对不同大豆品种的耐盐能力进行综合评价,筛选出相对耐盐的大豆品种,从而为盐碱地有效利用和大豆耐盐性评价提供理论依据.采用水培方法,用80 mmol·L^-1混合盐(NaCl:Na2SO4的摩尔比为9:1)溶液,对18份大豆品种的幼苗(25 d苗龄)进行盐胁迫.处理7 d后,对株高、茎粗、叶面积、主根长、茎干重、叶干重、根干重、叶片超氧化物歧化酶(SOD)活性、叶片游离脯氨酸含量和叶片丙二醛(MDA)含量等指标进行测定,通过主成分分析法和模糊数学隶属函数法对18个大豆品种进行耐盐性综合评价,并进行聚类分析.结果表明,经主成分分析,叶面积、主根长和丙二醛含量的负荷量最大,可作为衡量大豆耐盐性与耐盐品种筛选的主要指标.利用模糊数学隶属函数法对不同品种大豆进行了耐盐性排序,品种间耐盐性表现出明显差异;聚类分析结果显示,18份大豆品种可以被分为4大类,其中,东农42等5个品种为耐盐品种,东农60和雅布力为耐盐性较好的品种,吉育299等9个品种为中度耐盐碱性品种冻农63和吉育256为盐敏感品种. The saline tolerance of different soybean varieties was evaluated to screen out soybean varieties with relative salt tolerance,providing a theoretical basis for the utilization of soybean in saline-alkali land.Under hydroponic conditions,25-day-old seedlings of eighteen soybean varieties were treated with water containing 80mmol·L^-1 mixed salt(NaCl and Na2SO4 with the molar ratio of 9:1).Cluster analysis,the indexes for plant height,stem diameter,leaf area,main root length,stem dry weight,leaf dry weight,root dry weight,leaf SOD activity,leaf free proline content and leaf MDA content were measured after 7 days of stress treatment.The factor loadings of leaf area,main root length and leaf MDA content indexes were the largest by principal component analysis,and which could be used as the main identification indicators for the selection of salt tolerant soybean varieties.The salt tolerance of different soybean varieties was ranked by the subordinate function method of fuzzy mathematics.The salt tolerance of different soybean varieties showed obvious differences.Finally,throuyh cluster analysis,eighteen soybean varieties showed significant differences in salt tolerance,and they could be divided into four categories.Among them,5 soybean varieties including Dongnong 42 were salt tolerant.Dongnong 60 and Yabuli were better in salt tolerance.9 soybean varieties including Jiyu 299 were medium saline tolerant varieties,and two,Jiyu 256 and Dongnong 63,were saline sensitive.
