摘要
目的 :揭示蟾酥注射液 (ChansuInjection ,CHS)体外抑制淋巴瘤U937细胞增殖的机制。方法 :采用MTT法检测CHS对淋巴瘤U937细胞增殖的影响 ;采用Giemsa染色和透射电镜技术观察CHS处理的U937细胞形态学和超微结构变化 ;采用琼脂糖凝胶电泳检测CHS处理的U937细胞的DNA片段化。结果 :CHS 2 4h对淋巴瘤U937细胞增殖出现抑制 (P <0 .0 5)。CHS处理的U937细胞光镜和电镜出现核着边、核碎片和凋亡小体 ;CHS处理的U937细胞DNA电泳出现DNA梯状图谱。与对照组比 ,2 .2 5× 1 0 - 1 μg·mL- 1 CHS作用 ,2 4 ,48,72h后凋亡指数 (AI)升高 (P <0 .0 5) ,2 .2 5 ,2 .2 5× 1 0 - 1 ,2 .2 5× 1 0 - 2 μg·mL- 1 CHS作用 48h的U937细胞AI升高 (P <0 .0 5)。结论 :CHS蟾酥注射液体外抑制淋巴瘤U937细胞增殖 ,并且诱导其凋亡。
Objective: To investigate the mechanism of inhibition of lymphoma cell proliferation by Chansu Injection(CHS) (venenum Bufonis) in vitro . Methods: MTT assay was used to detect the effect of CHS on proliferation of lymphoma cell line U937. Giemsa stain, light microscope and transmission electron microscope were used to observe morphological and ultrastructural changes of lymphoma cell U937 treated by CHS. DNA gel electrophoresis was applied to detect DNA fragmentation of U937 cell treated by CHS. Results: The proliferation of U937 cell was inhibited after treatment of CHS for 24 hr( P <0.05). U937 cells treated with 2.25×10 -1 μg·mL -1 . CHS for 48 hr showed morphological and ultrastructural features changes such as in aggregation of chromation at the nuclear membrane, cell condensation and the formation of apoptotic bodies. U937 cells treated with CHS showed classical DNA ladder pattern. Apoptosis indexes(AI) of U937 cells 48 hr after treatment with CHS 2.25,2.25×10 -1 ,2.25×10 -2 μg·mL -1 were higher than those treated with control solution( P <0.05). AI of U937 cells that treated with CHS 2.25×10 -1 μg·mL -1 for 24,48,72 hr were higher than that of the control( P <0.05). Conclusion: CHS can induce lymphoma cell U937 apoptosis, which may be the mechanism of inhibition of lymphoma cell proliferation by CHS in vitro.
