摘要
本研究采用秋水仙素、8-羟基喹啉、秋水仙素和8-羟基喹啉1:1混合溶液,设计不同的时间梯度对美国流苏(Chionanthus virginicus)的组培苗根尖进行预处理,以获取可进行核型分析的高质量染色体制片。结果显示,美国流苏根尖经过3种试剂预处理后,均可获得分散良好、可计数且形态正常的染色体制片。秋水仙素最适预处理时间为2 h;8-羟基喹啉的最适预处理时间为3 h;秋水仙素和8-羟基喹啉混合溶液(1:1)的最适预处理时间为3 h。染色体核型分析表明:美国流苏的核型公式为2n=2x=46=36m+8sm+2msat;核型不对称系数为57.26%,为1B核型。本研究为美国流苏的染色体研究提供理论依据。
In this study,1:1 combination of colchicine,8-hydroxyquinoline,colchicine and 8-hydroxyquinoline were used to design different time gradients to pretreat the root tip of tissue culture seedlings of American tassel(Chionanthus virginicus).High quality chromosome preparation for karyotyping was obtained by the above method.The results showed that the American tassel root tip could be obtained by dispersing three kinds of reagents,and then the well-distributed,countable and normal morphological chromosomes were obtained.The optimal pretreatment time of colchicine was 2 h;whereas the optimum pretreatment time of 8-hydroxyquinoline colchicine and 8-hydroxyquinoline mixed solution(1:1)was 3 h,respectively.Karyotype analysis showed that the karyotype formula of American tassel was 2n=2x=46=38m+8sm+2msat;the karyotype asymmetry coefficient was 57.26%,which was 1B karyotype.This study would provide a theoretical basis for the study of chromosomes of American tassels.
