卵泡抑素样蛋白5基因在鹅肥肝形成中表达调控的研究 被引量:2

摘要 试验旨在探讨卵泡抑素样蛋白5(FSTL5)基因与鹅(Anser Cygnoides)肥肝形成的关系。试验选取30只健康、体重基本一致的70日龄朗德鹅,随机分为填饲组(15只)和对照组(15只)。在填饲到82日龄(填饲12 d)和94日龄(填饲24 d)时屠宰取样。此外,分离培养鹅原代肝细胞,并用葡萄糖、胰岛素和不同脂肪酸进行处理。结果显示:填饲后期肝脏和腹脂中FSTL5基因的表达量显著低于对照组(P<0.05),而胸肌中的表达量显著高于对照组(P<0.05)。在鹅原代肝细胞中,100 nmol/L胰岛素或0.25 mmol/L亚油酸能诱导FSTL5的表达(P<0.05)。综上所述,肝脏、脂肪与肌肉组织中的FSTL5表达在鹅肥肝形成后期受到显著影响,脂肪肝形成相关因子特别是胰岛素和亚油酸可诱导FSTL5的表达。研究结果为进一步研究FSTL5基因在鹅肥肝形成过程中的作用奠定了基础。 The aim of this study was to investigate the relationship between follistatin-like 5(FSTL5) gene and the formation of fatty liver in geese(Anser Cygnoides). A total of 30 healthy 70-day-old Langdes geese with similar weight were selected and randomly divided into overfed group(15) and control group(15). The geese were killed at 82 days and94 days of age. After sacrifice, tissues were immediately harvested from the geese. In addition, Goose primary hepatocytes were isolated from Landes goose embryos and treated with glucose, insulin and fatty acids. The results showed that the FSTL5 expression in liver and abdominal fat was significantly lower than that in control group at late stage of overfeeding(24 d)(P<0.05), while the expression in pectoral muscle was significantly higher than that in control group(P<0.05). In goose primary hepatocytes, 100 nmoL/L insulin or 0.25 mmoL/L linoleic acid could induce the expression of FSTL5(P<0.05). In conclusion, the expression of FSTL5 in liver, fat and muscle tissues was affected in the late stage of fatty liver formation in geese. Insulin and linoleic acid could induce the expression of FSTL5. This study laid a foundation for further study on the role of FSTL5 gene in the formation of goose fatty liver.
