摘要
为明确齐齐哈尔市郊区西瓜繁育基地西瓜幼苗猝倒的发病原因及为西瓜苗期病害防治提供依据,从发病区采集病样进行病原鉴定.结果分离获得12个菌株,根据培养特征和形态学特征,确定分离到的菌株为尖孢镰刀菌(Fusaruim oxysporum);利用真菌通用引物ITS1/ITS4扩增菌株rDNA-ITS序列,GenBank数据库BLAST同源比对与F oxysporum一致性达99%.将分离到的菌株回接到健康西瓜幼苗上,植株表现出与田间相似发病症状.基于形态学、分子生物学及致病性检测,最终确定齐齐哈尔市郊区育苗基地西瓜猝倒病的病原菌为Fusaruim oxysporum.
Watermelon seedlings with damping off symptom was found in Qiqihar suburban breeding base. In order to identify the cause of disease, samples were collected from the diseased area. A total of 12 strains were isolated and identified as Fusaruim oxysporum according to the characteristics of culture and morphology. The sequence of rDNA-ITS was amplified using primers ITS1/ITS4, and BLAST homologous alignment in GenBank database was 99% consistent with that of F. oxysporum The isolated strains were inoculated to healthy watermelon seedlings and the plants showed symptoms similar to those in the field. Based on morphology, molecular biology and pathogenicity detection, Fusaruim oxysporum was identified as the pathogen of watermelon damping off in Qiqihar city suburban breeding base.
