摘要
目的:研究从连翘叶中分离纯化高纯度连翘苷元的制备方法,为连翘苷元的规模化制备提供实验依据。方法:以浸膏中连翘苷元含量为考察指标,采用正交试验,确定水提最优工艺,提取液乙醇沉淀后,乙酸乙酯搅拌萃取、水反洗得到粗品。对粗品进行甲醇结晶,得高纯度的连翘苷元。结果:经水提醇沉萃取可得到纯度>75.0%的连翘苷元粗品,再经甲醇结晶,即可得纯度>99.0%的连翘苷元单体。结论:该工艺操作简单、成本低、效率高,可用于规模化制备连翘苷元。
Objective: To study the method of separating and purifying high purity forsythia glycogen from Forsythia suspensa leaves, thus to provide experimental basis for the large-scale preparation of forsythia glycogen. Methods: The content of forsythia glycogen in the extract was taken as the index, and the optimal process of water extraction was determined by orthogonal test. After the extraction solution was precipitated by ethanol, ethyl acetate was mixed and extracted, and the crude product was obtained by water backwash. High purity forsythia glycogen was obtained by crystallizing methanol in crude products. Results: After water extraction and alcohol precipitation extraction, the crude product of forsythia glycogen with purity above 75.0% could be obtained, and then through methanol crystallization, the purity could come up to more than 99.0%. Conclusion: The preparation technology is simple with low cost but high efficiency, which can be used as large-scale process to prepare forsythia glycogen.
