摘要
建立了超高效液相色谱-高分辨飞行时间质谱法对液态乳中的糠氨酸质量浓度进行测定。样品加入浓度为10.8 mol/L盐酸溶液后,进行氮气保护处理,于110℃条件下进行酸水解反应。反应后用纯水定容,离心,过滤。在优化的色谱及质谱条件下,采用ESI和APCI复合源和正离子扫描方式进行数据采集。糠氨酸标准曲线在10.0~1000μg/L范围内呈良好线性关系,相关系数r为0.9997,平均回收率为84.0%~86.2%,方法检出限为250μg/kg,相对标准偏差为1.08%~4.01%。该方法具有操作简便、高效快速、高回收率、灵敏度高和重现性好等优点,可用于实际测定液态乳中糠氨酸的质量浓度。
An ultra performance liquid chromatography-high resolution quadrupole time of flight mass spectrometer(UPLC-Q-TOF-MS)method for the determination of furosine in liquid milk was developed. Samples were hydrolyzed by 10.8 mol/L hydrochloric acid under 110 ℃ after Nitrogen protection treatment. Finally detected under the optimized chromatographic and mass spectrometer conditions, data were acquisited with ESI and APCI composite sources and positive ion mode. The calibration curves of furosine showed good linearity within the concentrations of 10~1000 μg/L(r>0.9997). The average recoveries of three spike levels were ranged 84.0%~86.2%, while the limits of detection were 250 μg/kg and the relative standard deviations(RSDs) were ranged 1.08%~4.01%. This method can be used to actually determine the content of furosine in liquid milk.
