褪黑素对脑缺血/再灌注脑损伤保护作用及机制 被引量:6

Protective effect of melatonin on brain injury of cerebral ischemia/reperfusion and its mechanism

摘要 目的探讨褪黑素对脑缺血/再灌注大鼠脑损伤的保护作用及机制。方法将90只大鼠根据随机数字法分为对照组、模型组和褪黑素组,每组30只。测定大鼠脑梗死体积和神经行为学评分,采用蛋白质印迹法(Western blot)法测定脑组织C/EBP同源蛋白(CHOP)、磷酸化胰腺内质网激酶(p-PERK)、葡萄糖调控蛋白78(GRP78)、裂解的半胱氨酰天冬氨酸特异性蛋白酶-3(cleaved Caspase-3)蛋白水平,采用反转录-聚合酶链反应(RT-PCR)测定大鼠脑组织CHOP、PERK、GRP78mRNA水平。应用SPSS20.0统计软件分析,计量资料以均值±标准差(Mean±SD)表示,组间比较采用t检验,3组之间比较采用方差分析,组内两两比较采用LSD检验。结果褪黑素组大鼠脑梗死体积比例[(17.83±1.46)%]和神经功能缺损评分[(1.34±0.22)分]均低于模型组[(44.25±2.92)%,(3.12±0.46)分],差异有统计学意义(t=30.280、18.472,P<0.05),差异有统计学意义。与对照组比较,模型组和褪黑素组大鼠脑组织CHOP、p-PERK、GRP78、cleaved Caspase-3蛋白水平升高(F=22.879、47.469、25.788、75.459,P<0.05),差异有统计学意义;与模型组比较,褪黑素组大鼠脑组织CHOP、p-PERK、GRP78、cleaved Caspase-3蛋白水平降低(t=5.134、6.274、5.618、4.382,P<0.05),差异有统计学意义。与对照组比较,模型组和褪黑素组大鼠脑组织CHOP、PERK、GRP78 mRNA水平升高(F=65.940、23.848、16.277,P<0.05),差异有统计学意义;与模型组比较,褪黑素组大鼠脑组织CHOP、PERK、GRP78mRNA水平降低(t=7.463、5.164、4.026,P<0.05),差异有统计学意义。结论褪黑素脑缺血/再灌注大鼠的脑组织具有保护作用,其机制可能通过抑制脑组织内质网应激发挥脑保护作用。 Objective To investigate the protective effect of melatonin on brain injury in rats with cerebral ischemia/reperfusion and its mechanism. Methods Ninety rats were divided into control group, model group and melatonin group accorded to random number method, thirty rats in each group. The cerebral infarction volume and neurobehavioral score of rats were determined. The C/EBP homologous protein (CHOP), phosphorylated pancreatic endoplasmic reticulase (p-PERK), glucose-regulated protein 78 (GRP78) and cleaved caspase-3 protein levels were determined by Western blotting analysis. The levels of CHOP, PERK and GRP78 mRNA in rat brain tissue were determined by reverse transcription-polymerase chain reaction (RT-PCR). Used t test and analysis of variance. Results The cerebral infarction volume [(17.83±1.46)%] and neurological deficit scores [(1.34±0.22) scores] of the melatonin group were lower than those of the model group [(44.25±2.92)%,(3.12±0.46) scores](t=30.280, 18.472, P<0.05). Compared with the control group, the levels of CHOP, p-PERK, GRP78 and cleaved caspase-3 protein in the brain tissue of the model group and the melatonin group were increased (F=22.879, 47.469, 25.788, P<0.05). Compared with the model group, the levels of CHOP, p-PERK, GRP78 and cleaved caspase-3 protein in the brain tissue of the melatonin group were decreased (t=5.134, 6.274, 5.618、4.382, P<0.05). Compared with the control group, the levels of CHOP, PERK and GRP78 mRNA in the brain tissue of the model group and melatonin group were increased (F=65.940, 23.848, 16.277, P<0.05). Compared with the model group, the levels of CHOP, PERK and GRP78 mRNA in the brain tissue of melatonin group were decreased (t=7.463, 5.164, 4.026, P<0.05). Conclusion Melatonin has a protective effect on brain tissue of rats with cerebral ischemia/reperfusion, and its mechanism may be that melatonin exerts brain protection by inhibiting endoplasmic reticulum stress in brain tissue.
