日粮中添加迷迭香及维生素E、维生素C和大豆油的组合对高温期蛋鸡不同组织HSP70、LYZ基因表达量的影响 被引量:13

Effects of combination adding rosemary,vitamin E,vitamin C and soybean oil to diet on the amount of expression of HSP70and LYZ genes in different tissues of laying hens in high temperature period

摘要 为研究高温期日粮中添加迷迭香、维生素E、维生素C和大豆油组合对蛋鸡热应激蛋白(HSP70)、溶菌酶(LYZ)基因表达量的影响,本试验选用41周龄京粉蛋鸡360只,随机分为3个处理组,每组3个重复,每个重复40只鸡。Ⅲ为对照组,饲喂基础日粮,Ⅰ组在基础日粮之上添加迷迭香草粉(0.6%),Ⅱ组在基础日粮之上添加维生素E(300 mg/kg)、维生素C(200 mg/kg)和大豆油(2%)(E_3C_2D_2组合)。试验结束当天,采集3个处理组各6只鸡的肝脏、心脏、脾脏、肺脏、胸腺、肾脏、小肠、腺胃、卵巢、峡部、膨大部和子宫的组织样品,测定各组织HSP70和LYZ基因的表达量。结果显示,Ⅰ、Ⅱ组蛋鸡心脏、肺脏、卵巢和子宫部位HSP70基因的表达量极显著低于对照组(P<0.01);脾脏、腺胃HSP70基因的表达量极显著高于对照组(P<0.01);肾脏HSP70基因的表达量显著低于对照组(P<0.05);肝脏HSP70基因的表达量显著低于对照组(P<0.05)。Ⅰ、Ⅱ组蛋鸡肝脏、心脏、脾脏、肺脏、小肠、腺胃、卵巢和峡部部位LYZ基因的表达量极显著高于对照组(P<0.01);Ⅱ组子宫、肾脏LYZ基因的表达量极显著高于迷迭香组(Ⅰ组)和对照组(P<0.01),而胸腺LYZ基因的表达量极显著低于对照组(P<0.01)。结果表明,日粮中添加0.6%迷迭香和E_3C_2D_2组合可降低心脏、肺脏、肾脏、卵巢和子宫部位HSP70基因的表达量,增加肝脏、心脏、脾脏、肺脏、小肠、腺胃、卵巢和峡部部位LYZ基因的表达量,从而缓解热应激对蛋鸡生产性能造成的影响,延长鸡蛋的保存时间。 In order to study the effect of adding rosemary,vitamin E,C and soybean oil todiet on the amount of expression of heat stress protein(HSP70) and lysozyme(LYZ) genes of tissues in high-temperature period,a total of 360 laying hens of Jingfen of 41 weeks-old were randomly divided into 3 treatment groups with 3 replicates in each group and 40 chickens each.Treatment group Ⅲ was as the control group,fed with the basal diet.Treatment groupⅠand Ⅱ were the experimental group.Rosemary herb powder(0.6%),vitamin E(300 mg/kg),vitamin C(200 mg/kg) and soybean oil(2%)(E3C2D2 combination) were added the basal diet.On the last day of the experiment,tissue samples of liver,heart,spleen,lung,thymus,kidney,small intestine,glandular stomach,ovary,isthmus,enlarged part,and uterus of 6 chickens in each of the three treatment groups were collected,and the amount of expression of HSP70 and LYZ genes in each tissue were measured.The results showed that the amount of expression of HSP70 gene in heart,lung,ovary and uterus of experimental group with 0.6% rosemary and E3C2D2 combination was significantly lower than that in the control group(P<0.01),significantly higher than that in the control group(P <0.01)in spleen and glandular stomach,significantly lower than that in the control group(P<0.05)in the kidney,significantly lower than that in the control group(P<0.05)in the liver.The amount of expression of LYZ gene in the liver,heart,spleen,lung,small intestine,glandular stomach,ovary and isthmus of experimental group with 0.6% rosemary and E3C2D2 was significantly higher than that in the control group(P<0.01),significantly higher than that in the rosemary group and the control group(P<0.01)in uterus and kidney,significantly lower than that in the control group(P<0.01)in the thymus.Adding to diet with 0.6%rosemary and E3C2D2 combination reduced the amount of expression HSP70 gene expression in heart,lung,kidney,ovary and uterus,and increased the amount of expression LYZ gene in liver,heart,spleen,lung,small intestine,glandular stomach,ovary and isthmus,mitigating the impact of heat stress on the performance of laying hens and extending the egg preservation time.
