Optimization of Extraction Technology of Polysaccharide in Eleocharis tuberosa Peel by Inner Ebullition with Box-Behnken Experimental Design

摘要 为改善荸荠皮多糖提取效果,在单因素试验的基础上,采用Box-Behnken试验设计优化内部沸腾法提取荸荠皮多糖的提取条件。结果表明,在乙醇浓度和液料比分别为80%和20∶1(mL/g)时,最佳工艺条件为:pH 5,温度80℃,时间20 min,该条件下多糖得率的理论预测值为19.55%,此条件下验证试验的平均得率为为20.07%,相对标准偏差(relative standard deviation,RSD)为3.45%,与理论预测值的相对误差为2.66%。试验结果为荸荠皮的综合利用提供了理论基础。 To improve extraction effect of polysaccharide in Eleocharis tuberosa peel, at the basis of single-factors experiments, the Box-Behnken experimental design was used to optimize the extraction conditions, in which polysaccharide was extracted from Eleocharis tuberosa peel by inner ebullition method. The results showed that when the ethanol concentration and liquid-solid ratio were 80 % and 20 ∶ 1 (mL/g), respectively, the optimal technological condition were pH of 5, temperature of 80 ℃, and time of 20 min, in which the theoretical prediction value of polysaccharide yield was 19.55 %. Under the optimal conditions, the average yield of polysaccharide in verification experiment was 20.07 %, relative standard deviation(RSD) was 3.45 %, which relative error was 2.66 % for the theoretical predication value. Theoretical basis was provided for comprehensive utilization of Eleocharis tuberosa peel by the experimental results.
