肝癌微环境中FOXP3^+调节性T细胞对肿瘤免疫抑制作用的研究 被引量:10

摘要 目的探讨肝细胞癌组织浸润FOXP3^+调节性T细胞及其与肿瘤免疫的关系。方法对100例肝细胞癌组织和癌旁组织采用免疫组化方法检测FOXP3^+调节性T细胞和CD8^+细胞数量,并计算FOXP3^+T/CD8^+T比值进行分析,用荧光定量PCR检测肝细胞癌组织和癌旁组织免疫抑制细胞因子IL-10的表达量。结果肝癌及癌旁组织中FOXP3^+T细胞阳性单个高倍视野平均数分别为26.7±6.2和53.3±8.7,P=0.0074;肝癌及癌旁组织中CD8^+T细胞阳性单个高倍视野平均数分别为51.3±7.1和125.0±10.3,P=0.0005。肝癌及癌旁组织中FOXP3^+T/CD8^+T值分别为0.519±0.06和0.422±0.02,P=0.017。肝细胞癌组织免疫抑制细胞因子IL-10的mRNA表达量比癌旁组织更高(P<0.001)。结论FOXP3^+调节性T细胞通过抑制CD8^+T细胞增殖,分泌免疫抑制细胞因子IL-10抑制肝癌的局部免疫,导致肿瘤细胞增殖。 Objective To investigate the infiltration of FOXP3^+regulatory T cells in hepatocellular carcinoma and its relationship with tumor immunity.Methods The number of FOXP3^+regulatory T cells and CD8^+cells were detected by immunohistochemistry in 100 cases of hepatocellular carcinoma and paracancerous tissues.The ratio of FOXP3^+T/CD8^+T was calculated and analyzed.The expression of immunosuppressive cytokine IL-10 in hepatocellular carcinoma and paracancerous tissues was detected by quantitative PCR.Results The mean number of FOXP3^+T cell positive single high power visual field in hepatocellular carcinoma and adjacent tissues were 26.7±6.2 and 53.3±8.7,P=0.0074,respectively.The mean number of CD8^+T cell positive single high power visual field in hepatocellular carcinoma and adjacent tissues were 51.3±7.1 and 125.0±10.3,P=0.0005,with significant difference.The values of FOXP3^+T/CD8^+T in hepatocellular carcinoma and adjacent tissues were 0.519±0.06 and 0.422±0.02,respectively,P=0.017,with significant difference.The expression of IL-10 in hepatocellular carcinoma tissues was higher than that in adjacent tissues(P<0.001).Conclusion FOXP3^+regulatory T cells inhibit the local immunity of tumors by inhibiting the proliferation of CD8+T cells and secreting immunosuppressive cytokine IL-10.
