摘要
以胡桃楸成熟胚为外植体,研究了外植体消毒及培养预处理(低温、暗培养)对胡桃楸不定芽诱导的影响。结果表明:外植体消毒处理中以紫外线直射5 min+0.1%HgCl_28 min处理效果好于其他处理,染菌率为13.45%,褐化率为22.78%。培养预处理中暗培养随着时间延长愈伤组织诱导率增加。低温处理是随着时间延长呈现抛物线形式,先升后降,低温14 d的愈伤组织诱导率最高,为42.71%。经过预处理后,在低温处理14 d的不定芽诱导率最高,为42.8%。随着种子贮存年限的延长,胡桃楸发芽率、愈伤组织诱导率、不定芽诱导率越低。
We studied the effect of disinfection and culture pretreatment(low temperature and dark culture)on adventitious bud induction for the mature embryo of Juglans mandshurica Maxim.The treatment of disinfection for UV direct 5 min+0.1%HgCl 2 8 min is better than other treatments.The rate of bacterial infection is 13.45%and browning rate was 22.78%.The induction rate of the callus is increased in the dark culture and is parboiled in the low temperature treatment as time goes on.The induction rate of the callus is the highest for 14 d at low temperature with 42.71%.Adventitious bud induction rate reaches the highest(42.8%)at the low temperature treatment of 14 d.The lower germination rate,callus induction rate and adventitious bud induction rate happens with the prolongation of seed storage life.
