摘要
目的探讨骨肉瘤患者在不同新辅助化疗方案实施前后多药耐药相关蛋白(MRP)在mRNA以及蛋白水平中的表达变化。方法选取2016年1月至2017年1月收治的骨肉瘤患者48例,根据辅助化疗方案不同分为2组,检测患者化疗前后患者MRP表达情况。结果同一化疗方案以及不同化疗方案治疗前后,患者MRP表达无明显区别(P> 0. 05);化疗前后患者Cyelin D1阳性表达同预后无显著相关性(P> 0. 05),化疗前患者Bel-2阳性表达同预后无显著相关性(P> 0. 05),化疗后患者阳性表达同预后显著相关(P <0. 05),阳性表达越低预后质量越好。结论新辅助化疗方案对骨肉瘤患者MRP无明显影响,耐药性主要受肿瘤组织的影响,临床上检测MRP表达可以为化疗方案的制定提供指导。
Objective To investigate the changes of expression levels of multidrug resistance associated protein(MRP)at mRNA and protein levels in patients with osteosarcoma before and after neoadjuvant chemotherapy.Methods A total of 48 patients with osteosarcoma who were admitted and treated in our hospital from January 2016 to January 2017 were enrolled in the study,who were divided into two groups according to the different adjuvant chemotherapy protocols.The expression levels of MRP before and after chemotherapy were detected and compared between the two groups.Results There were no significant differences in the expression levels of MRP at the same chemotherapy protocol and in different chemotherapy protocols before and after treatment(P >0.05).There was no significant correlation between the positive expression of CyelinD1 and patient’s prognosis before and after chemotherapy(P >0.05).There was no significant correlation between the positive expression of Bel-2 and patient’s prognosis before chemotherapy(P >0.05),however,there was close correlation between the positive expression of Bel-2 and patient’s prognosis after chemotherapy(P<0.05).Conclusion The neoadjuvant chemotherapy has no obvious effects on MRP in patients with osteosarcoma,and drug resistance is mainly affected by tumor tissues,and the clinical detection of MRP expression can provide guidance for the chemotherapy plan.
