摘要
目的探究鼻咽上皮细胞特异性蛋白1(nasopharyngeal epithelium specific protein 1,NESG1)在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达及临床意义。方法收集临床及随访资料完整的150例非小细胞肺癌患者的癌组织及相应癌旁正常肺组织并制成组织芯片,利用免疫组织化学技术Envision二步法检测NESG1在肺癌及癌旁组织中的蛋白表达情况。结果 150例非小细胞肺癌患者的癌组织及相应癌旁正常肺组织免疫组化结果示,NSCLC组织中NESG1阳性表达率为33. 33%(50/150),显著高于癌旁正常组织的6. 47%(9/139),2组差异有统计学意义(χ2=32. 03,P=0. 00)。NESG1表达与淋巴结转移、TNM分期和肿瘤大小等临床病理变量密切相关(P <0. 05),而与患者的年龄、性别、组织类型及病理分级差异无统计学意义(P> 0. 05)。Kaplan-Meier生存分析显示NESG1阴性表达组患者的生存时间明显高于NESG1阳性表达组,二者间差异有统计学意义(P <0. 05)。多因素Cox比例风险模型分析显示NESG1是NSCLC患者预后的一项独立因素。结论 NESG1在NSCLC表达上调与肿瘤的发生发展密切相关,是一项独立的预后因素。它在NSCLC中的具体角色、分子机制还需要进一步研究。
Objective To explore the expression and clinical significance of nasopharyngeal epithelium specific protein 1( NESG1) in patients with non-small cell lung cancer( NSCLC). Methods A total of 150 patients with NSCLC were enrolled in the study. The specimens of cancer tissues and corresponding normal lung tissues were collected and made into tissue chip. The immunohistochemical technique was used to detect the expression of NESG1 in 150 cases of NSCLC cancer tissues and corresponding normal lung tissue. The relationship between the NESG1 expression and clinical pathological characteristics as well as the prognosis of patients with NSCLC was statistically analyzed. Results The positive expression rate of NESG1 in NSCLC tissue was 33. 33%( 50/150),which was significanty higherr than that [6. 47%( 9/139) ] in canceradjacent normal tissues( P < 0. 01). Moreover the expression of NESG1 was closely correlated with tumor size,lymph node metastasis,clinical TNM staging( P < 0. 05),however,which was not correlated to patient’s age and sex,histological type and pathological grading( P > 0. 05). Kaplan-Meier survival analysis showed that the survical time in NESG1-negative group was significantly higher than that in NESG1-positive group( P < 0. 05). In addition multivariate analyses by Cox’s proportional hazard model showed that NESG1 was an independent prognostic factor of NSCLC. Conclusion NESG1 is an independent prognostic factor of NSCLC,and its expression in NSCLC is closely correlated with pathogenesis and development of NSCLC,however its molecular mechanism still needs to be researched further.
