摘要
目的探讨趋化因子CCL2对小鼠动脉血栓形成的影响。方法选取8~10周龄野生型C57雄性小鼠和CCL2^(-/-)雄性小鼠制备小鼠颈总动脉血栓模型。应用小动物超声探头连续监测小鼠颈总动脉血流变化,记录小鼠颈总动脉闭合时间。结果 CCL2^(-/-)小鼠血液中血小板和白细胞计数与野生型C57小鼠比较,差异无统计学意义(P> 0. 05)。CCL2^(-/-)小鼠鼠尾平均出血时间较野生型C57小鼠明显延长(P <0. 05)。CCL2^(-/-)小鼠血凝块的容积均较野生型C57小鼠大(P <0. 05)。CCL2^(-/-)小鼠颈总动脉平均闭合时间较野生型C57小鼠明显延长(P <0. 05)。结论趋化因子CCL2参与了小鼠动脉血栓形成。
Objective To investigate the role of chemokine CCL2 in mice arterial thrombosis.Methods Wild type C57 male mice and CCL2^-/-male mice aged 8-10 weeks were selected to prepare a mouse model of common carotid artery thrombosis.The changes of common carotid artery blood flow were continuously monitored by small animal ultrasound probe,and the closing time of common carotid artery was recorded.Results There was no statistically significant difference in platelet and white blood cell counts between CCL2^-/-mice and wild-type C57 mice(P>0.05).The mean tail bleeding time of CCL2^-/-mice was significantly longer than that of wild-type C57 mice(P<0.05).The blood clot volume of CCL2^-/-mice was larger than that of wild-type C57 mice(P<0.05).The mean closing time of common carotid artery of CCL2^-/-mice was significantly longer than that of wild-type C57 mice(P<0.05).Conclusion CCL2 participates in arterial thrombosis.
