摘要
目的:通过对COPD大鼠基因表达谱芯片的分析,探讨全真一气汤抑制COPD大鼠肺损伤的作用机制。方法:用气管注入内毒素LPS+烟熏法制造COPD大鼠模型,分为模型组9只、中药组9只,中药组予中药全真一气汤浓缩液灌胃治疗,60d后随机各组取3只大鼠取材,应用大鼠基因表达谱芯片分析两组大鼠肺组织基因差异,并利用IPA整合分析系统分析差异基因与copd的关系。结果:全真一气汤干预的中药组大鼠IL-8信号通路被显著抑制,Z-score为-2.333,其中下调基因PIK3R3(FC=-2.1091385,logFC=-1.0766538)、HMOX1(FC=-2.1376743,logFC=-1.096042)、GNAS(FC=-2.4846587,log FC=-1.3130478)、PLD3(FC=-2.4485795,log FC=-1.2919451)、ITGAM(FC=-2.500478,log FC=-1.322204)、CXCR1(FC=-2.5748348,log FC=-1.3644799)、LASP1(FC=-2.2364473,log FC=-1.1612087)、CR2(FC=-4.4339395,log FC=-2.1485891)。结论:全真一气汤可能通过调控IL-8信号通路中相关基因表达,从而抑制了COPD大鼠的肺损伤,为中医中药治疗慢性肺部疾病提供理论依据。
Objective:According to analysis gene expression profiles of COPD rats,to explore the mechanism of the Quanzhen Yiqi decoction(全真一气汤)on inhibiting lung injury in COPD rats.Methods:COPD rat models were made by endotoxin LPS plus fumigation.COPD rats were divided into the model group(9 rats)and TCM medicine group(9 rats).The TCM medicine group was treated with the Quanzhen Yiqi decoction by gastric perfusion.After 60 days,3 rats were randomly selected from each group.The gene differences between groups were analyzed by gene expression profiles.The relationship between differentially expressed genes and COPD was analyzed by IPA integration.Results:IL-8 signaling pathway was significantly inhibited in the TCM medicine group,with Z-score of-2.333,including to(FC=-2.1091385,logFC=-1.0766538),HMOX1(FC=-2.1376743,logFC=-1.096042),GNAS(FC=-2.4846587,logFC=-1.3130478),PLD3(FC=-2.4485795,logFC=-1.2919451),ITGAM(FC=-2.500478,logFC=-1.322204),CXCR1(FC=-2.5748348,logFC=-1.3644799),LASP1(FC=-2.2364473,logFC=-1.1612087),CR2(FC=-4.4339395,logFC=-2.1485891).Conclusion:The Quanzhen Yiqi decoction may inhibit lung injury in COPD rats by regulating the expression of related genes in IL-8 signaling pathway;it could provide theoretical basis for treating chronic lung diseases in TCM.
