芍药苷抑制佐剂性关节炎大鼠FLS异常增殖 被引量:7

Paeoniflorin suppresses the FLS abnormal proliferation in adjuvant arthritis rats

摘要 目的:研究芍药苷(PF)对佐剂性关节炎(AA)大鼠关节滑膜成纤维样滑膜细胞(FLS)异常增殖的影响。方法:采用完全弗氏佐剂足跖注射法制备AA大鼠,造模后第28天股动脉放血处死大鼠,分离培养FLS,大鼠足爪肿胀评分法检测PF灌胃治疗对AA大鼠的治疗作用,ELISA检测PF对AA大鼠FLS细胞因子IL-8表达的影响,MTT检测PF加入FLS培养液对AA大鼠FLS增殖的影响,Real time q PCR检测PF加药对AA大鼠FLS促凋亡基因Bax、凋亡因子Caspase 3、抗凋亡基因Bcl-2、RA病理基因Fibronectin表达的影响。结果:PF显著抑制AA大鼠足爪肿胀程度,PF加入FLS培养液后对FLS增殖具有一定的抑制作用,对AA大鼠FLS中Bax、Caspase 3表达具有显著上调作用,对Bcl-2、Fibronectin、IL-8表达具有显著抑制作用。结论:PF可能通过抑制AA大鼠FLS异常增殖抑制AA大鼠病理发展。 Objective:To investigate the role of Paeoniflorin(PF)in the abnormal proliferation of fibroblast-like synoviocytes(FLS)in adjuvant arthritis(AA)rats.Methods:AA rats were prepared by the complete Freund′s adjuvant,and the rats were sacrificed at28days after modeling,then the FLS were cultured.The effects of PF gavage treatment on the AA rats were detected by rat paw edema score,the PF impact on cytokines IL-8 expression in AA rats was detected by ELISA.The effect of PF dosing on the FLS proliferation was detected by MTT,the effects of PF dosing on the expression of pro-apoptotic gene Bax,apoptosis factor Caspase3,anti-apoptotic gene Bcl-2,RA pathology gene fibronectin were detected by real time qPCR.Results:PF significantly inhibited the rat paw swelling,suppressed the FLS abnromal proliferation,significantly up-regulated the expression of Bax and Caspase3,inhibited the expression of Bcl-2,fibronectin and IL-8.Conclusion:PF may inhibit the AA pathological development by inhibiting the FLS abnormal proliferation.
