摘要
叶绿素是光合作用主要色素,叶绿素含量的高低和动态变化直接影响水稻穗部籽粒产量的形成。以典型直立穗粳稻品系沈农0530-9和大穗型高产籼稻北陆129杂交衍生的F2和F2∶3群体为试材,对不同生育时期叶绿素含量与穗部性状进行QTL(quantitative trait loci)分析。与直立穗粳稻沈农0530-9相比,高产籼稻北陆129不同生育时期均具有较高的叶绿素含量,且生育中后期叶绿素降解较为缓慢;不同生育时期亲本和遗传群体的叶绿素含量存在明显动态变化特征,叶绿素含量与穗部性状存在明显相关性,其中抽穗期和成熟期的叶绿素含量与穗部性状关系最密切。共检测到26和25个控制不同生育时期叶绿素含量和穗部性状相关的QTL,分布在1~11号染色体上;叶绿素含量与穗部性状相关QTL位点具有明显的遗传重叠和相对独立遗传特性,分布于第1、2、3、6和7号染色体上的QTL簇在控制叶绿素含量和穗部性状方面具有明显的遗传重叠效应,5个染色体区域包含22个相关的位点,占QTL总数44.0%,其他QTL位点遗传相对独立,其中位于第3染色体R3M10~RM6676区域和第6染色体RM587~RM19660区域具有明显的遗传多效性,不同生育时期的叶绿素含量和穗粒性状的遗传重叠效应较为明显;第2染色体RM1347~RM6247区域的qCJ2a、qCH2a、qCM2、qCD2、qTGW2、qGN2、qPW2和qSSR2是一个新的影响叶绿素含量和穗粒性状的多效性主效QTL,暂将其命名为qCHY2,该区域来自沈农0530-9的增效等位基因对生育中后期叶绿素含量具有明显的降低效应,具有明显降低穗粒数和籽粒充实性状的效应。研究结果进一步阐明了叶绿素含量和穗部性状间存在相互独立与相互依赖两种遗传关系,也为相关主效QTL簇的精细定位和功能解析奠定基础。
Chlorophyll content is one of the most important photosynthetic parameters.The content and dynamic changes of chlorophyll have a significant effect on the yield of rice.In this study,F2 and F2∶3 populations,derived from a cross of the typical erect panicle japonica rice Shennong 0530-9 and the large panicle type indica rice Habataki,were used to identify QTLs for chlorophyll content and panicle traits at different stages of rice.Compared with the Shennong 0530-9,Habataki had higher chlorophyll content and slower chlorophyll degradation in the middle and late growing stages.The chlorophyll content of parents and genetic populations at different growth stages had obvious dynamic changes,there was significant correlation between panicle traits and chlorophyll content under different growth stages,especially at the heading and maturity stage of chlorophyll content and yield traits had significant relationship,which chlorophyll content was the most closely related to panicle traits.Twenty-six and twenty-five QTLs for chlorophyll content and panicle traits were detected on the chromosome 1-11,these QTLs had obvious genetic overlap and independent genetic characteristics.These pleiotropic genetic overlap effect QTLs were clustered on the chromosome 1,2,3,6 and 7,five major QTL cluster regions contain 22 related QTL,accounting for 44.0%of the total number of QTL,and then others QTL had independent genetic characteristics.Two major QTL cluster were located on the chromosome 3(R3M10~RM6676)and 6(RM587~RM19660)had obvious genetic overlap effect,and then chlorophyll content and the genetic overlap effect of panicle and seed traits were obvious at different growth stages.In addition,a novel major cluster QTL qCHY2(qCJ2a,qCH2a,qCM2,qCD2,qTGW2,qGN2,qPW2 and qSSR2)was located on the chromosome 2(RM1347-RM6247)for chlorophyll content and yield,the active allele from Shennong 0530-9 had a positive effect on chlorophyll content in the middle and late stages of growth,and the grain number and seed filling were significantly decreased.These results were important for more comprehensively clarifying the independent and interdependent genetic relationships between chlorophyll content and panicle traits,also would help us with fine mapping of the major QTL clusters and provide genetic mechanism of theoretical and information.
