摘要
目的预测潜在的γ-和β-珠蛋白表达调控因子,为珠蛋白基因表达调控研究提供新的思路。方法从GEO数据库中下载γ-和β-珠蛋白基因差异表达相关的两组RNA-seq数据,利用分化各时期共同存在的不同组织来源细胞间的差异表达基因及其预测出的上游调控因子,通过STRING数据库、Centiscape软件进行潜在调控因子的预测。结果除预测到BCL11A、NF-E2、YY1和GATA1等已知的γ-和β-珠蛋白基因表达调控因子外,从两组数据共同预测出14个潜在调控因子。结论分析预测出的包括MAPK1、TNF和IFNG等潜在的调控因子可能调控γ-和β-珠蛋白基因表达,需要近一步的分子生物学实验证实。
Objective To predict potential regulators ofγ-andβ-globin gene.Methods The RNA-seq data of differential expression betweenγ-andβ-globin were analyzed to screen the differential expressed genes and IPA was used to explore the upstream regulators.Prediction ofγ-andβ-globin gene regulators was conducted by STRING database and Centiscape software.Results Known globin gene regulators such as BCL11A,NF-E2,YY1,GATA1 were predicted in the protein-protein interaction network,and 14 potential regulators were predicted from both two sets of data.Conclusions MAPK1,TNF,IFN and so on regulators being predicted may regulateγ-andβ-globin gene expression.It requires further molecular biology experiments to confirm.
