醒脑静注射液对β淀粉样蛋白25-35诱导的SH-SY5Y细胞氧化损伤的影响 被引量:1

Effects of Xingnaojing Injection on Oxidative Damage Induced by Aβ25-35 in SH-SY5Y Cells

摘要 目的观察醒脑静注射液对β-淀粉样蛋白25-35(Aβ25-35)诱导的SH-SY5Y细胞氧化损伤的影响,探讨其相关作用机制。方法用不同浓度醒脑静(5、10、20μL/m L)预处理SH-SY5Y细胞3 h,随后以25μmol/L Aβ25-35处理SH-SY5Y细胞24 h制备阿尔茨海默病体外模型。实验细胞分为正常对照组、模型组和醒脑静低、中、高剂量组,采用流式细胞术检测细胞凋亡率,DCFH-DA荧光探针检测细胞内活性氧(ROS)含量,酶标法检测细胞内丙二醛(MDA)、还原型谷胱甘肽(GSH)、超氧化物歧化酶(SOD)水平。结果与正常对照组比较,模型组中细胞内ROS含量明显增加,MDA水平明显升高,GSH、SOD水平明显下降(P<0.05);与模型组比较,不同浓度醒脑静预处理组细胞凋亡率降低,细胞内ROS含量明显降低,MDA水平明显下降,GSH、SOD水平显著升高(P<0.05)。结论醒脑静注射液预处理可抑制Aβ诱导的细胞氧化损伤,减少细胞凋亡,发挥细胞保护作用。 Objective To investigate the effects of Xingnaojing Injection on oxidative damage induced byβ-amyloid protein 25-35(Aβ25-35)in SH-SY5Y cells;To discuss its mechanism of action.Methods SH-SY5Y cells with different concentrations of Xingnaojing injection(5,10,20μL/mL)were pretreated for 3 hours,and then the external model of alzheimer's disease was established by treating SH-SY5Y cells with 25μmol/L Aβ25-35 for 24 hours.The experimental cell were randomly divided into normal control group,model group,and Xingnaojing low-,medium-,and high-dose groups.The apoptotic rate was measured by flow cytometry and intracellular reactive oxygen species(ROS)was measured by DCFH-DA fluorescent probe.The expressionsof MDA,GSH and SOD in the cells were measured by enzyme standard instrument.Results Compared with the normal control group,the apoptotic rate of the model group was significantly increased;the content of ROS increased and the MDA level increased(P<0.05);GSH and SOD were significantly decreased(P<0.05).Compared with the model group,the apoptotic rate of the pretreatment group was significantly lower than that of the model group;the content of ROS decreased and the MDA level decreased(P<0.05);GSH and SOD were significantly increased(P<0.05).Conclusion Xingjingjing Injection pretreatment can inhibit the cell oxidative damage and the cell apoptosis induced by Aβ25-35,which can play a cytoprotective effect.
